头孢曲松钠预处理对局灶性脑缺血大鼠缺血周围脑组织白介素1β和白介素6表达的影响 被引量:4

摘要 目的探讨头孢曲松钠预处理对局灶性脑缺血大鼠缺血周围脑组织白介素1β(IL-1β)和白介素6(IL-6)表达的影响。方法选取24只健康雄性SD大鼠,随机分为假手术组、模型组和预处理组,各8只。预处理组大鼠于术前每日腹腔注射头孢曲松钠200 mg/kg,共5 d;假手术组和模型组大鼠分别给予等量的0.9%氯化钠注射液腹腔注射,给药方法及疗程与预处理组相同。然后制备大脑中动脉闭塞(MCAO)模型,而假手术组大鼠术中不插入线栓,其余过程与模型组及预处理组相同。术后24 h内进行神经功能评分,并用免疫组化法检测大鼠脑缺血周围组织IL-1β、IL-6阳性表达率。结果 MCAO术后,假手术组、模型组、预处理组大鼠神经功能评分〔0、(2.5±0.9)、(1.5±0.8)分〕及脑缺血周围组织IL-1β阳性表达率〔(7.3±1.3)%、(82.7±12.4)%、(52.7±9.9)%〕、IL-6阳性表达率〔(6.8±2.6)%、(69.3±10.2)%、(50.4±9.8)%〕比较,差异均有统计学意义(P<0.05);其中模型组、预处理组大鼠神经功能评分及脑缺血周围组织IL-1β、IL-6阳性表达率均高于假手术组(P<0.01),而预处理组大鼠上述3个指标均低于模型组(P<0.01)。结论头孢曲松钠能提高局灶性脑缺血大鼠神经功能,可能是通过抑制脑组织中炎性递质IL-1β和IL-6的表达,从而减少炎性损伤来起作用。 Objective To explore the effect of ceftriaxone pretreatment on the expression of IL - 1β and IL - 6 around the ischemic tissue in rats with focal cerebral isehemia. Methods 24 healthy male Sprague - Dawley rats were randomly divided into sham operation group, model group and pretreatment group with each group 8 rats. The pretreatment group was given intraper- itoneal injection of 200 mg/kg ceftriaxone sodium for five days; The sham operation group and model group were given intraperito- neal injection of sodium chloride, with the same dose and treating course. The model of middle cerebral artery of occlusion (MCAO) was then established, in which the middle cerebral artery occlusion was not included for the sham operation group. The neurological function score was given within 24 hours after operation and the expression of IL - 1β and IL - 6 in hippocampus was determined by immunohistochemistry. Results After MCAO, the neurological scores in the sham operation group, model group and pretreatment group were 0, (2. 5 ± 0. 9 ) and ( 1.5 ± 0. 8 ) respectively, and the positive expression rates of IL - 1β around the ischemia tissue were (7.3 ± 1.3 ) % , ( 82.7 ± 12.4) % and ( 52.7 ± 9.9) % , and the rates for IL - 6 were ( 6. 8± 2.6 ) % , ( 69. 3 ± 10. 2) % and ( 50. 4 ± 9. 8 ) %, all showing statistically significant differences ( P 〈 0. 05 ) . The neurological scores and positive expression rates of IL - 113 and IL - 6 in the model group and pretreatment group were higher than the sham operation group (P 〈 0. 01 ), and the above indicators were lower in pretreatment group than in model group (P 〈 0.01 ) . Conclusion Ceftriaxone sodium can improve the neurological function of rats with focal cerebral ischemia probably by inhibiting the expression of inflammatory cytokines IL - 1β and IL - 6, thus reducing inflammatory injury.
