大鼠牙髓干细胞、外胚间充质干细胞的细胞表型的实验研究

摘要 目的检测组织特异性分化标记在大鼠牙髓干细胞(DPSC)、外胚间充质干细胞(EMSC)及骨髓间充质干细胞(BMSC)中的表达,探讨三者细胞表型的异同。方法通过免疫组化检测大鼠DPSC、EMSC、BMSC三种细胞的表型;RT-PCR检测DPSC、EMSC、BMSC的DSPP mRNA表达。结果免疫组化结果显示,三种细胞对于不同细胞的表面标志有不同程度的表达;RT-PCR显示,DSPP的mRNA在大鼠DPSC、EMSC中不表达,在BMSC中弱表达。结论 DPSC与EMSC的细胞表型具有一定的相关性和延续性。 Objective To investigate the expression of special differentiation marker in dental pulp stem cell (DPSC), ectomesenchymal stem cell (EMSC) and bone mesenchymal stem cell (BMSC), to compare the difference of the phenotype among the three kinds of cell. Methods The phenotype of rat DPSC, EMSC and BMSC was described by immunohistochemistry. The mRNA expression of DSPP in rat DPSC, EMSC and BMSC was detected by RT-PCR. Results Immunohistochemistry showed different degree of expression of cell surface markers in the three kinds of cells. The DSPP mRNA was not detected in rat DPSC and EMSC, but little detected in BMSC by RT-PCR. Conclusion The special cell phenotypes of rat DPSCs and EMSC were in possession of certain relevance and continuity.
