摘要
目的观察亚低温联合丁苯肽对大鼠脑缺血再灌注损伤的保护作用。方法制备SD大鼠MCAO模型,梗死2h后再灌注,并分为假手术组、模型组、丁苯肽组及亚低温联合丁苯肽组(联合组),再灌注24h后进行神经功能缺损评分检测脑组织超氧化物歧化酶(SOD)、丙二醛(MDA)浓度、病理变化及大脑缺血面积并比较。结果 (1)丁苯肽组与联合组的神经功能缺损评分明显低于模型组,且联合组的神经功能缺损评分明显低于丁苯肽组(P<0.01)。(2)与假手术组相比,模型组SOD活性降低,MDA含量增加(P<0.01);与模型组相比,丁苯肽组与联合组SOD活性增高,MDA含量降低(P<0.01),且联合组与丁苯肽组相比差异有统计学意义(P<0.01)。(3)模型组、丁苯肽组及联合组病理学检查均较假手术组差,但联合组好于丁苯肽组好于模型组(P<0.01)。(4)模型组、丁苯肽组及联合组脑缺血面积较假手术组增大,而联合组<丁苯肽组<模型组,差异均有统计学意义(P<0.01)。结论亚低温联合丁苯肽对脑缺血再灌注损伤有较好的保护作用。
Objective To observe the effect of mild hypothermla combined with Dinsbentai soft capsule against eerebrd ischemia reperfusion injury in rats. Methods 64 SD rats were divided randomly into 4 groups(n= 16). sham operation group, normal saline(NS) group, Dlngbentat soft capsule group and mild hypothermia eotnbtned with Dingbentat soft capsule group, Nerve function deficit scores levels of brain tissue SOD enzyme(SOD) and malondialdehyde(MDA)and brain ischemia area were detected after 24 hours with reperfusion, The morphoiogtc ehanges were observed by microscope,Results (1) Compared with NS group, neural function defect score was significantly lower in Dingbental soft capsule group and the combination 8fotlp than that in NS group, and in mild hypothermta combined with Dingbentat soft capsule group the neural function defect score was significantly lower than that of Dingbentat sob capsule group(P〈0.01), (2) Compared with sham operation group, the aetivhy of SOD deereased and the content of MDA increased in NS group (P〈0. 01) 1 and compared to the NN group, the activity of SOD increased in Dtngbelatai soft cup,ale group and combination group+ the content of MDA decreased (P〈0.01) and there were significant differences between eombtned group and Dingbental soft capsule group(P〈0. 011, (3) The results of pathological examination in NS group, Dtnghentai soft e^psule group and mild hyprathernia combined with Dingbental soft capsule group rule group were different compared with sham operation group, but the re+ultra tn mild hypothertttla combined with Dingbental soft capsule were better than Dingbentai soft capsule group, and better than that of Ng group(P〈0. 01), (4) Cerebral ischemla area in NS group, Dingbental soft capsule group and mild hypothermia combined with Singbental soft capsule group Increased compared with sham operation group (P〈0. 01). Conclusion Mild hypothermia combined with Dingbental soft capsule offered offered protection against cerebral tschemla reperfusion injury in rifts,
