摘要 目的 从中药蒲公英花中分离纯化总黄酮.方法 用超声波助提法得到蒲公英花黄酮粗提液,采用D-101 大孔吸附树脂对其进行纯化,并研究影响动态吸附与解吸的各种因素.结果 最佳分离纯化条件为:在D-101 树脂柱中,上样液浓度为1.238 mg/ml,pH 值为3,上样量为2 树脂床体积倍数(BV),室温下以流速2.0BV/h 吸附饱和,用4 BV 的50%~70% 乙醇以2 BV/h 的流速洗脱,在此条件下,测得黄酮纯度为92.56%,吸附率为80.65%,解吸率为53.44%,回收率为42.8%.结论 在所确定的条件下,D-101 大孔吸附树脂分离纯化蒲公英花总黄酮效果良好,为蒲公英花总黄酮的开发利用提供了新途径. Objective To separate and purify the total flavonoids from Taraxaeum mongolicum infloreseences (TMI). Methods The crude flavonoid extracts of TMI was prepared by ultrasonic extraction and purified with D-101 resin, which entailed the analysis on various factors contributing to dynamic absorption and desorption. Results The optimal technical conditions for purification was to supplement the maeroporous D-101 resins with 2 BV of crude flavonoid samples (pH: 3.0) at a velocity of 1.238 mg/ml at room temperature till saturation (2.0 BV/h), followed by an elution with 4 BV 50%-70% ethanol at the rate of 2.0 BV/h. This gave a purity of 92.56%, an adsorption rate of 80.65%, a desorption rate of 53.44% and a recovery rate of 42.8%. Conclusion The D-101 macroporous adsorption resin confers a satisfactory capacity for purifying the total flavonoids from TMI under an optimal condition, which of-fers a new approach for developing and utilizing the total flavonoids.
