摘要
石榴皮中含有丰富的单宁类物质,具有良好的抑菌活性。以丙酮为提取溶剂,采用超声波辅助法对石榴皮单宁进行提取,并用Amberlite XAD-16大孔吸附树脂纯化,经高效液相色谱对其组分进行分析后,采用琼脂稀释法做最小抑菌浓度实验,结果表明:石榴皮提取物的主要成分为安石榴苷,纯化后质量浓度由12.3%提高到64.2%;石榴皮粗提物对金黄色葡萄球菌、单核细胞增生李斯特氏菌、沙门氏菌和大肠杆菌的最小抑菌浓度分别为0.312、10、0.625mg/mL和大于10mg/mL,纯化物对四种菌的最小抑菌浓度分别为0.156、5、0.625、10mg/mL。
Pomegranate peel is rich in tannins which have a good antibacterial activity. Pomegranate peel tannins were extracted by acetone in ultrasonic-assisted extraction method. After that they were purified by Amberlite XAD-16 macroporous resin. Then high performance liquid chromatography(HPLC) analysis was taken to determine the components and agar dilution method was adopted to test the minimal inhibitory concentration(MIC). The results indicated that the concentration of punicalagin,the main component of pomegranate peel extract,increased from 12.3% to 64.2% after purification. The MICs of pomegranate peel crude extract on Staphylococcus aureus,Listeria monocytogenes,Salmonella and E.coli were 0.312,10,0.625mg/mL and larger than 10mg/mL,while the MICs of purified extract for these four kinds of bacteria were 0.156,5,0.625 and 10mg/mL respectively.
