摘要 目的 观察CUG连接蛋白1(CUGBP1)在肝癌中的表达及其对肝癌细胞增殖能力的影响.方法 免疫组织化学法检测20例正常肝组织标本及60例肝癌组织标本中CUGBP1的表达,并探讨CUGBP1表达与肝癌患者临床病理特征的关系;同时利用小干扰RNA (siRNA)对肝癌细胞中CUGBP1表达进行下调,通过细胞计数试剂盒(CCK-8)法检测CUGBP1下调后细胞的增殖能力.结果 CUGBP1在肝癌中的阳性表达率为66.7%,明显高于正常组织(0%),差异有统计学意义(P<0.05).同时,CUGBP1表达与分化程度及淋巴结转移有关(P<0.05);CUGBP1表达下调后,肝癌细胞的增殖能力明显下降(P<0.05).结论 CUGBP1表达与肝癌的形成、分化及转移密切相关,其表达下调可明显抑制细胞增殖. Objective To investigate the expression of CUG-binding protein 1 (CUGBP1) in hepatocellular carcinoma,and its effect on cell proliferation.Methods CUGBP1 expression was detected in 20 cases of normal liver and 60 cases of hepatocellular carcinoma by using immunohistochemistry,and the relationship between CUGBP1 expression and clinicopathologic features was assessed.Cell Counting kit-8 (CCK-8) assay was used to examine cell proliferation when CUGBP1 expression was knocked down by small interfering RNA (siRNA).Results The positive rate of CUGBP1 in hepatocellular carcinoma tissues was 66.7%,significantly higher than those that in normal liver tissues (0%,P 〈 0.05).CUGBP1 expression was obviously associated with tumor differentiation and lymph node metastasis in epatocellular carcinoma (P 〈 0.05).After CUGBP1 downregulation,the ability of cell proliferation was significantly decreased (P 〈 0.05).Conclusion CUGBP1 expression is closely associated with the formation,differentiation and lymph node metastasis of hepatocellular carcinoma,and CUGBP1 downregulation can obviously inhibit cell proliferation.
