摘要
目的 观察ALX4基因、增生性息肉蛋白基因(HPP1)和胰岛素样生长因子结合蛋白3基因(IGFBP3)同时出现甲基化对胃癌预后的影响.方法 应用Methylight聚合酶链反应(PCR)分析200例胃癌患者ALX4、HPP1和IGFBP3的甲基化状态.结果 多因素分析显示,3个基因联合检测同时出现甲基化表型可作为独立预后因素[24.158(7.248 -31.611),P<0.01];与患者淋巴结转移(P<0.01)和TNM分期相关(P<0.01).结论 ALX4、HPP1和IGFBP3同时出现高甲基化表型与临床病理学特征相关,与患者预后较差有关.
Objective To explore the influence of simultaneous methylation of aristaless-like 4 (ALX4),hybrid-type peptide pheromone precursor 1 (HPP1) and insulin-like growth factor-binding protein 3 (IGFBP3) on the prognosis of gastric cancer.Methods Methylation status of the ALX4,HPP1 and IGFBP3 was assessed in 200 patients with gastric cancer using methylight polymerase chain reaction (PCR).Results Multiple factor analysis revealed that the detection of simultaneous methylation expression of the three genes could be used as an independent prognosis factor [24.158 (7.248-31.611),P 〈0.01],and correlated with lymph node metabasis (P 〈 0.01) and TNM staging (P 〈 0.01).Conclusion The simultaneous hypermethylation expression of ALX4,HPP1 and IGFBP3 was correlated with the clinicopathological characteristics,and is associated with poor prognosis of patients with gastric cancer.
