摘要
目的 观察乙二醛酶1(GLO1)在胶质瘤中的表达及其对胶质瘤C6细胞增殖迁移及侵袭能力的影响.方法 应用免疫组织化学法检测48例胶质瘤标本中GLO1的表达,同时采用小干扰RNA (siRNA)对C6细胞中GLO1的表达进行下调,分别采用Transwell小室及噻唑蓝(MTT)检测GLO1下调对细胞增殖迁移及侵袭能力的影响.结果 GLO1在胶质瘤中的阳性表达率为60.4%,其表达与病理分级有关(P<0.05);GLO1表达下调后,C6细胞的增殖迁移及侵袭能力均明显下降(P<0.05).结论 GLO1的表达与胶质瘤的形成密切相关,其表达下调可明显抑制细胞的增殖迁移及侵袭能力.
Objective To detect the expression of glyoxalase 1 (GLO1) in gliomas and to investigate its role in migration,proliferation and invasive ability of C6 cell line.Methods GLO1 expression was examined in 48 cases of glioma tissues by using immunohistochemistry.After downregulation of GLO1 expression in C6 cells using small interfering RNA (siRNA) interference fragment,the ability of migration,proliferation and invasion was evaluated by transwell assay and methyl thiazol tetrazolium (MTT) assay,respectively.Results The positive rate of GLO1 in gliomas was 60.4%,and the GLO1 expression was related to the pathological grade (P 〈 0.05).GLO1 downregulation significantly inhibited migration,proliferation and invasion of C6 cells (P 〈 0.05).Conclusion GLO1 expression is closely associated with the formation of glioma,and its downregulation can obviously inhibit migration,proliferation and invasion.
