Notch1在人食管鳞状细胞癌中的表达及意义

Decreased Notch1 expression in esophageal squamous cell carcinoma

目的 探讨食管鳞癌组织中Notch1的表达水平及其意义.方法 收集30例食管鳞癌组织和食管正常黏膜上皮组织,采用免疫组织化学法、免疫组织荧光和Western blot法分析食管鳞癌组织及食管正常黏膜组织Notch1蛋白表达水平的变化.结果 免疫组织化学染色显示食管鳞癌组织中Notch1蛋白表达水平（3.56±1.45）低于食管正常黏膜组织中染色指数（12.45 ±3.12）,差异有统计学意义（P＜0.01）.免疫组织荧光和Western blot结果亦显示食管鳞癌组织中Notch1蛋白表达低于食管正常黏膜组织.结论 Notch1蛋白在食管鳞癌组织中低表达,在正常食管黏膜组织中高表达,提示Notch1基因在食管鳞癌的发生发展过程中可能起负调控作用.

Objective The aim of this study was to determine whether Notch1 expression was altered in esophageal squamous cell carcinoma （ESCC） compared with normal epithelium （NORMAL）.Methods We examined Notch1 expression in ESCC and compared to the NORMAL by immunohistochemical staining and Western blotting.Results By immunohistochemical staining Notch1 protein expression levels are decreased in ESCC （12.45 ± 3.12） compared to the NORMAL （3.56 ± 1.45）,P 〈 0.01.Meanwhile,We got the similar results by immunofluorescence histochemistry and immunoblot analysis.Conclusion Notch1 may act as an important molecule in the esophageal SCC regulation.
