摘要 目的采用Box-behnken设计-响应面法优化平菇中多糖的提取工艺。方法以超声波辅助纤维素酶的提取方法,考察纤维素酶解温度、酶解时间、酶用量和超声时间对提取率的影响,应用SAS 9.1软件模拟得到二次响应回归方程,并通过典型分析得到最佳的提取条件。结果最佳提取条件为:酶解温度52℃,酶解时间57min,酶用量0.016 5g,超声时间5min,该条件下提取率为26.09%。结论 Box-behnken响应面设计优化平菇多糖的提取工艺可行,为进一步工业化提取提供了依据。 Objective To optimize the extraction procedure of polysaccharides from Pleurotus ostreatus by using Box-behnken design and response surface method. Methods Polysaccharides from Pleurotus ostreatus were extracted by ultrasonic-assisted cellulase hy- drolysis. The multiple quadratic regression describing the relationship between the extraction rate of polysaccharides and parame- ters such as,enzymatic hydrolysis temperature,enzymatic hydrolysis time,enzymatic quantities and hyperacoustic time to raw ma- terial was established by using a 4-factor,3-1evel Box-behnken statistical design. Response surface and contour plots with the ex- traction rate of polysaccharides as the response were drawn based on the model. Results The optimal conditions of extraction were determined as follows: enzymatic hydrolysis temperature 52 ℃,enzymatic hydrolysis time 57 min,enzymatic quantities 0. 016 5 g and ultrasonic time 5 min. Under such conditions, the extraction rate was 26.09 %. Conclusions The optimized extraction process is feasible and has provided a basis for further industrialized extraction.
