摘要
目的 :建立一种莫海林分子量及其分布测定的质控方法。方法 :采用高效凝胶渗透色谱法 (HPGPC) ,色谱柱为ShodexAsahipakGF 5 10HQ (7 6mm× 30 0mm) ,以已知分子量的葡聚糖 (Dextran)为标样 ,0 2mol·L-1硫酸钠 (稀硫酸调至pH 5 0 )为流动相 ;柱温 35℃ ;流速 0 5mL·min-1;示差折光检测器。结果 :利用GPC软件处理 ,得到莫海林的数均、重均分子量和分子量累积重量分布曲线。结论 :本文所建立的HPGPC法简便、快速、重现性好 。
Objective:To set up a method for the determination of molecular weight and molecular weight distribution for the quality control of moherin.Methods:The average molecular weight(Mn,Mw)and molecular weight distribution curves of moherin were determined by high performance gel permeation chromatography(HPGPC)on Shodex Asahipak GF-510 column with 0 2 mol·L -1 (pH 5 0)sodium sulfate solution as mobile phase using dextrans with known molecular weight as calibration standards.The flow rate was 0 5 mL·min -1 .The column temperature was 35 ℃ and RI detector was used.Results:The Mn,Mw and molecular weight distribution curves (cumulative weight fraction distribution curves)were obtained by GPC software from the calibration curves which were made by standard dextrans.Conclusion:The method is simple and quick with good reproducibility ( RSD <1%) and suitable for the control of the medicine.
