高效液相色谱法测定何首乌及其炮制品中蒽醌类成分的含量

HPLC Determination of Anthraquinone in Radix Polygoni Multiflori and Radix Polygoni Multiflori Preparata

目的 :采用反相高效液相色谱法 ,对 9个产地的生首乌和 9个产地的制首乌商品药材中水解前、后蒽醌类成分的含量进行测定。方法 :采用InertsilODS - 3柱 ,以甲醇 - 0 1%磷酸 (85∶15 )为流动相 ,检测波长 2 5 4nm。结果 :大黄素、大黄素甲醚、大黄酸、大黄酚、芦荟大黄素在水解前的加样回收率分别为 98 1% ,98 8% ,99 2 % ,99 9% ,99 8% ,精密度 (RSD )分别为 0 8% ,2 1% ,0 9% ,1 9% ,1 5 % ,n=5 ;水解后的加样回收率分别为 96 2 % ,96 1% ,98 3% ,97 6 % ,97 4% ,精密度 (RSD) 分别为 0 7% ,0 6 % ,1 8% ,1 6 % ,2 2 % ,n =5。结论 :各产地的生首乌和制首乌商品药材中蒽醌类苷元主要为大黄素和大黄素甲醚 ,某些产地的药材中另含微量的大黄酸、大黄酚和芦荟大黄素 ,其含量随产地差异较大。

Objective:To determine the contents of anthraquinone before and after hydrolysis in the commercial medicinal materials of Radix Polygoni Multiflori and Radix Polygoni Multiflori Preparata collected from different producing areas respectively by RP-HPLC.Method:This method used a column of Inertsil ODS-3 and methanol-0 1% phosphoric acid solution(85∶15)as the mobile phase,the detection wavelength was 254 nm.Results:The method recoveries of emodin,physcion,rhein,chrysophanol and aloe-emodin before hydrolysis were 98 1%,98 8%,99 2%,99 9%,99 8% respectively with corresponding RSD of 0 8%,2 1%,0 9%,1 9%,1 5%(n=5) and after hydrolysis were 96 2%,96 1%,98 3%,97 6%,97 4% respectively with corresponding RSD of 0 7%,0 6%,1 8%,1 6%,2 2%(n=5).Conclusion:Anthraquinone contained in the samples were mainly emodin and physcion ,besides,trace rhein,chrysophanol and aloe-emodin were detected in some samples.The contents varied greatly in the samples from the different producing areas.
