摘要
目的探讨维甲酸(RA)致神经管畸形(NTDs)中牛磺酸对神经细胞凋亡的影响。方法建立NTDs模型并给予牛磺酸拮抗,观察神经细胞中Caspase-3、Caspase-8表达的变化。结果各组Caspase-3、Caspase-8蛋白的表达在E11d达高峰,以后逐渐下降。RA组神经上皮细胞Caspase-3、Caspase-8蛋白在E9d^E13d的表达均高于对照组及牛磺酸组,差异有统计学意义(P<0.05)。各组Caspase-3mRNA、Caspase-8mRNA在E10d达到高峰,以后逐渐下降。RA组Caspase-3mRNA、Caspase-8mRNA在E9d^E13d的表达均高于对照组及牛磺酸组,差异有统计学意义(P<0.05)。结论牛磺酸可以拮抗RA引起的神经上皮细胞过度凋亡,其机理可能是通过下调Caspase-3、Caspase-8的表达实现的。
Objective To explore the effects of taurine on nerve cell apoptosis in neural tube defects (NTDs) caused by retinoic acid(RA). Methods NTDs model in mouse embryo was set up and taurine was given for antagonists to observe the changes in the expressions of Caspase-3 and Caspase-8 in nerve cells. Results The protein expressions of Caspase-3 and Caspase-8 in each group reached the peak in the Elld , then gradually declined. The expressions of Caspase-3 and Caspase-8 of neuroepithelial cells in the RA group in the Egd-E13d were significantly higher than those in the control group and the taurine group( P d0.05). The expressions of Caspase-3mRNA and Caspase-8 mRNA in each group reached the peak in the E10d, and then gradually declined. The expressions of Caspase-3 mRNA and Caspase-8 mRNA of neuro- epithelial cells in RA group in the E9d - E13d were significantly higher than those in the control group and the taurine group( P d0.05). Conclusion Taurine can antagonize the excessive apoptosis of neural epi thelial cells in NTDs caused by RA. Its molecular mechanism may be got through the down-regulation of the expressions of Caspase-3 and Caspase-8.
