摘要
目的: 采用近红外光谱透射法快速测定栀子逆流提取过程中总苷含量。 方法: 以UV分析值作为参照,采用NIR透射法结合偏最小二乘法建立总苷含量的快速在线测定方法。采样方式为透射采集,采集区间4 000-10 000 cm-1,分辨率8.0 cm-1,扫描数64次,empty门衰减,增益为1,温度(25±2) ℃,相对湿度45%-50%。 结果: 建立的栀子逆流提取液中总苷校正模型的相关系数、校正均方差分别为0.993,0.179。经外部验证,校正模型的预测均方差0.145,预测值和真实值间回归方程的相关系数达0.991。 结论: NIR可作为一种准确、快速的在线检测方法用于中药提取过程中有效成分的含量检测。
Objective: To develope a near-infrared quantitative method for rapid determination of total glycosides in countercurrent extracting process of Gardeniae Fructus. Method: UV was used as a reference method to determine the content of total glycosides in Gardeniae Fructus,a rapid on-line determination method of total geniposides was established by NIR transmission method and partial least squares method.Sampling method for transmission acquisition,collection interval 4 000-10 000 cm-1,resolution 8.0 cm-1,numner of scans 64 times,empty door attenuation,gain of 1,temperature at (25±2) ℃,relative humidity 45%-50%. Result: Correlation coefficients and calibration root-mean-square error of calibration model for total glycosides in countercurrent extract of Gardeniae Fructus were 0.993 and 0.179,respectively.By external validation,prodiction root-mean-square error of calibration model was 0.145,correlatiaon coefficient of regression equation between the actual value and the predicted value was 0.991. Conclusion: NIR could be used as a kind of accurate,rapid and on-line detection method for determination of active ingredients in extracting process of traditional Chinese medicine.
