Determination of Oleanolic Acid and Hederagenin in Yanyan Granule by HPLC-ELSD

目的： 建立测定咽炎颗粒中齐墩果酸与常春藤皂苷元的高效液相色谱-蒸发光散射含量测定方法。 方法： 采用高效液相色谱法，Cosmosil PAQ-C18（4.6 mm×250 mm，5 μm）色谱柱，甲醇（A）-水（B）流动相梯度洗脱，蒸发光散射检测，流速0.9 mL·min-1。 结果： 齐墩果酸的线性范围为1.321-5.285 μg（r=0.999 3），平均加样回收率为98.5%；常春藤皂苷元的线性范围为0.473 6-1.894 μg（r=0.999 2），平均加样回收率为100.2%。 结论： 该方法操作简便，结果准确可靠，可作为咽炎颗粒中齐墩果酸与常春藤皂苷元的含量测定方法。

Objective： To establish a method for the determination of both oleanolic acid and hederagenin in Yanyan granule by high performance liquid chromatography and evaporative light scattering detection （HPLC-ELSD）. Method： A HPLC-ELSD system was applied, using a Cosmosil PAQ-C18 （4.6 mm×250 mm,5 μm） column;mobile phase consisted of methanol （A）-water （B） under gradient elution;the flow rate was 0.9 mL·min-1. Result： As for oleanolic acid and hederagenin, the linear range was between 1.321-5.285 μg（r=0.999 3）and 0.473 6-1.894 μg（r=0.999 2）, with average recovery rate to be 98.5% and 100.2%, respectively. Conclusion： The method developed is simple, rapid with reliable results, which could be applied for the determination of oleanolic acid and hederagenin in Yanyan granule.
