二脱甲氧基姜黄素与阿霉素联合应用对HL-60/ADR的生长抑制作用

Inhibitory Effect of Combined Administration of Bisdemethoxy curcumin and Adriamycin on HL-60/ADR Cells

摘要 目的: 研究二脱甲氧基姜黄素(Bdmc)与阿霉素(ADR)联合应用对人白血病多药耐药细胞株 (HL-60/ADR)的生长抑制作用。 方法: HL-60/ADR细胞2×103/孔,1,2,4,8 μmol·L-1 Bdmc分别与0.4,0.8,1.6,3.2 μmol·L-1 ADR同时联合给药,48 h 后,MTT法测定两种药物的体外杀伤作用。6,12,24,48 μmol·L-1Bdmc分别与0.08,0.2,0.4,1.0 μmol·L-1ADR序贯联合给药,48 h 后,应用金氏公式进行序贯联合用药效果分析。 结果: 单独ADR 0.4,0.8 μmol·L-1时抑制率6.9%,11.8%,同时联合加8 μmol·L-1Bdmc时,抑制率达到94%以上。1-8 μmol·L-1 Bdmc和0.8-3.2 μmol·L-1ADR同时联合作用 HL-60/ADR细胞可产生单纯相加至增强的协同杀伤效果。序贯给药法中,先给Bdmc后给SDR结果为协同作用,先给ADR后给Bdmc实验结果为拮抗作用。 结论: Bdmc与ADR同时用药可产生协同作用,二者序贯联合用药仅产生单向协同作用。 Objective: To investigate the inhibitory effect of combined administration bisdemethoxy curcumin (Bdmc) and adriamycin(ADR) on multidrug resistance HL-60/ADR cells. Method: HL-60/ADR cells were simultaneously treated with Bdmc within concentrations (1,2,4,8 μmol·L-1) and ADR(0.4,0.8,1.6,3.2 μmol·L-1)48 h respectively, the cell density was 2×104/hole. The inhibitory effect of both drugs on HL-60/ADR cells in vitro was evaluated by MTT. Cells were reated sequentially administration Bdmc(6,12,24,48 μmol·L-1) and ADR(0.08,0.2,0.4,1.0 μmol·L-1). The Jin's formula was used to analyze the the inhibitory effect. Result: The inhibition is 6.9%,11.8% when treated only with ADR(0.4,0.8 μmol·L-1),while simultaneously treated with Bdmc(8 μmol·L-1) it was over 94%.In simultaneous administration Bdmc (1-8 μmol·L-1) combined with ADR(0.8-3.2 μmol·L-1) revealed a simple addition or potentiation effect. In addition, sequential administration of Bdmc followed by ADR had a synergistic effect;in contrast, sequential administration of ADR followed by Bdmc exhibited an antagonistic effect. Conclusion: Simultaneous administration of Bdmc and ADR reveals a synergistic effect and sequential administration of Bdmc and ADR exhibits an unidirectional synergistic effect.
