摘要
目的:探讨苗医验方四大血对大鼠佐剂性关节炎的作用及其机制。方法:建立类风湿性关节炎动物模型-佐剂性关节炎大鼠(adjuvant arthritis,AA),设立空白组、模型对照组、四大血高、中、低剂量组(按生药计分别是40,20,10 g·kg-1)、阳性对照组(雷公藤多苷片)。测量各组大鼠右后足跖肿胀度;大鼠滑膜细胞原代培养,取3代的传代培养细胞做实验。蛋白免疫印迹法(Western-blotting)检测各组大鼠滑膜细胞B细胞淋巴瘤/白血病2(Bcl-2)、Bcl-2相关x蛋白(Bax)蛋白质表达水平。结果:四大血有效抑制AA大鼠右足跖肿胀(P<0.05),且下调Bcl-2表达(P<0.05,P<0.01)、上调Bax表达(P<0.01),降低Bcl-2/Bax表达比值。结论:四大血拮抗AA病情发展,其作用机制可能与促进滑膜细胞凋亡,抑制滑膜细胞过度增生密切相关。
Objective: To discuss the effects of traditional Miao national herbs Sidaxue on rheumatoid arthritis and its mechanism. Method: An animal model of adjuvant arthritis (AA) was established, rats were divided into blank control group, model control group, high dose Sidaxue treatment group, middle dose Sidaxue treatment group, low dose Sidaxue treatment groups, positive control group(glucosidorum tripterygll totorum control group). The rat paw edemas of rats was measured;Synovial cells were primarily cultured, and the 3rd generations of subculture cell were adopted for experiment. Western-blotting method was adopted to detect the protein expression level of B cell lymphoma/leukemia-2(Bcl-2), Bcl-2 associated X protein(Bax). Result: Miao national herbs Sidaxue could inhibit the swelling of paw (P 〈 0.05);decrease the protein expression of Bcl-2 (P 〈 0.05,P 〈 0.01), increase the protein expression of Bax in Arthritis rats' synoviocyte(P 〈 0.01), and decrease the rate of Bcl-2/Bax. Conclusion: Miao national herbs Sidaxue can antagonize the development of AA, the mechanism of which may be related to promoting apoptosis of synoviocyte.
