柱切换-高效液相色谱法与荧光偏振免疫法快速监测苯巴比妥血药浓度的比较 被引量:2

Comparison on the methods of determination of serum concentration of phenobarbital by column switching-high performance liquid chromatography and fluorescene polarization immunoassay

摘要 目的:比较柱切换-高效液相色谱法和荧光偏振免疫法快速监测卡马西平血药浓度的结果相关性,评价柱切换-高效液相色谱法用于苯巴比妥治疗药物监测的可行性。方法:收集兰州军区兰州总医院门诊共52例癫痫患者血样,在同一天内分别用柱切换-高效液相色谱法和荧光偏振免疫法同时测定苯巴比妥血药浓度。以柱切换-高效液相色谱法测定值(X),荧光偏振免疫法测定值(Y)进行相关回归分析和配对t检验。结果:柱切换-高效液相色谱法和荧光偏振免疫法测定值之间相关性良好,回归方程为:Y=0.995 3 X+0.046 2(n=52,r=0.999 0);测定值两者之间差异无显著性(P>0.05)。结论:柱切换-高效液相色谱法灵敏、简单、方便、可靠,且显著降低监测成本,可适用于广大医院临床血药浓度的常规监测。 OBJECTIVE To compare the methods of determination of serum concentrations of phenobarbital by column switching high performance liquid chromatography (column switching-HPLC) and fluorescene polarization immunoassay (FPIA), and evaluate the feasibility of column switching HPLC for routine therapeutic drug monitoring. METHODS A total of 52 epileptic serum samples were determined by column switehing-HPLC and FPIA simultaneously. I.inear regression analy- sis was performed with column switching-HPLC results as x-axis and FPIA results as y-axis to evaluate their correlation . Paired t test was performed with these data. RESULTS There was a good correlation between column switching-HPLC and FPIA results. The linear equation was Y = 0. 995 3X + 0. 046 2(n = 52, r = 0. 999 0). The results determined by column switc hing HPI.C and FPIA showed no outstanding difference (n = 52, P)0. 05). CONCLUSION The method of column switching- HPLC was sensitive, simple, convenient and reliable, and could decrease monitor cost. It can he applied to routine monitoring of blood drug concentration in clinic of general hospitals.
