牙周膜干细胞移植促进兔牙槽骨再生的实验研究 被引量:4

摘要 目的:研究牙周膜干细胞(periodontal ligament stem cell,PDLSC)复合透明质酸钠凝胶载体(sodium hyaluronic acid gel,HAG)移植在兔下颌牙槽骨缺损区促进骨再生的效能与机制。方法 :拔除兔下颌磨牙,获取牙周膜组织做细胞培养;在兔下颌骨双尖牙区邻牙间隙的下方制备6 mm×6 mm×3 mm骨缺损,建立兔下颌牙槽骨骨缺损的动物模型,实验共分为4组,透明质酸钠凝胶复合牙周膜干细胞组(HAG+PDLSC组)、单纯牙周膜干细胞组(PDLSC组),透明质酸钠凝胶组(HAG组)和空白对照组。通过组织学检测、免疫组化染色和骨计量学研究分析各组修复效果。结果:透明质酸钠凝胶复合牙周膜干细胞组与其他各组相比,各指标差异均有显著性(P<0.05);单纯牙周膜干细胞组与空白组及单纯透明质酸钠凝胶组在成骨情况比较无明显统计学差异(P>0.05)。结论:单纯牙周膜干细胞移植没有明显促进成骨的性能,当透明质酸钠凝胶作为细胞载体,牙周膜干细胞可显著促进牙槽骨再生修复重建,该复合体具有良好的临床应用前景。 Objective To evaluate the osteogenic function of hyaluronangel combined with PDLSCs in rabbit mandibular alveolar defects. Method Mandibular alveolar defects surgically made in 24 rabbits were treated with HAG/PDLSCs in the experimental groups. Single periodontal ligament stem cells, single HAG carrier application were served as control groups. Nothing added to the animal defects were named as blank control. Histopathologic method, immune-histic staining, and histomorphometry were used to evaluate the osteogenic effects in the above groups. Result There was no significant difference between the control group and blank control group; However, significant difference was verified in the experimantal group. Conclusion It is verified that HAG/PDLSCs could promote osteogenesis in rabbit alveolar defect reconstruction. PDLSC has the positive functions in alveolar osteogenesis.
