摘要
目的:探讨CD4+CD25+Treg在牙周炎发生时的免疫调节作用。方法:选择35只成年SPF级Wistar大鼠,丝线结扎加高糖饮食建立大鼠牙周炎发生的动物模型,获得类似牙周炎自然病程的连续组织标本,病理切片确定病损,分别获取第4、8、12、16周的大鼠外周血,术前未结扎组对照。以流式细胞仪检测不同时段外周血中CD4+CD25+Treg的比例变化。结果:建立了Wistar大鼠牙周炎发生的动物模型,病理切片确定了牙周病损的存在。结果显示伴随牙周炎发生的牙槽骨的吸收逐渐增多。流式细胞仪结果显示结扎后第4、8、12、16周外周血中CD4+CD25+Treg占CD4+T细胞的比例为7.70%、9.12%、9.74%、10.31%,与术前比较差异均有统计学意义(P<0.05)。结论:外周血中CD4+CD25+Treg自牙周炎发生早期逐渐升高,牙周炎发生发展规律一致,从而证实CD4+CD25+Treg细胞参与介导了牙周炎发生的免疫调控。
Objective:To explore CD4+CD25+T cells in peripheral blood during the progression of experimental periodontitis. Method:35 adult SPF Wistar rats were selected during rat model of periodontitis occurs by means of high-sugar diet and silk ligature. Histological sections and peripheral blood of periodontitis tissue samples were obtained respectively at 4,8,12,16 weeks. Non-ligation group as control. The frequency of CD4+CD25+T cells in peripheral blood during the progression of experimental periodontitis was measured by fluorescence-activated cell sorter(FACS)method. Result:Wistar rat model was established to determine the existence of histology periodontal loss. The results showed that the absorption of alveolar bone associated with periodontitis occurs gradually increased. Flow cytometry showed that the frequency of CD4+CD25+T cells accounted for 7.70%,9.12%,9.74%, 10.31%at the 4,8,12,16 weeks after ligation compared with control group were statistically significant(P〈0.05).Conclusion:During the progression of experimental periodontitis,the percentage of CD4+CD25+Treg increase in peripheral blood. CD4+CD25+Treg cells may play regulatory roles in the progression.
