摘要 探讨了以DEAE Sepharose FF阴离子交换树脂为纯化介质的大豆植酸快速测定方法,主要对测定过程中植酸的纯化、消化方法进行了优化。在提取方法的基础上(料液比1∶50(质量体积比)),6 mL的树脂体积能够充分交换上样液中的植酸;用15 mL蒸馏水、15 mL 0.05 mol/L NaCl淋洗,能够较好达到除游离磷效果;植酸溶液经过消化炉300℃浓缩后,加入3 mL浓硫酸以400℃消化0.5 h,回收率达到90%以上。此方法的加标回收率达108.4%,3个大豆样品中植酸测定结果的相对标准偏差分别为4.91%、2.82%和1.24%。 Rapid determination method of phytic acid of soybean has been evaluated adopting of DEAE sepha- rose FF weak base anion - exchange resin, including purification, digestion of phytic acid. On the base of extraction method (50-fold volume (mL/g)), phytic acid of soybean was exchanged fully when resin volume is 6 mL; The free phosphorus of sample was removed fully when the column was washed with 15 mL distilled water and 15 mL 0.05 % mo]/L NaC1 respectively; Purified phytic acid was concentrated on 300℃ by digestion furnace, and on 400 % for 0.5 h. When 3 mL concentrated sulfuric acid added, the recovery was more than 90%. The of added phytic acid was up to 108.4% , the relative standard deviations of the method for phytic acid of 1,2 and 3 were 4.91%, 2.82% and 1.24% respectively.
