急性心肌梗塞患者红细胞磷脂过氧化氢谷胱甘肽过氧化物酶及Ca^(2+)-Mg^(2+)-ATP酶活性的变化

摘要 目的 :探讨急性心肌梗塞 (acute myocardial infarction,AMI)患者红细胞磷脂过氧化氢谷胱甘肽过氧化物酶 (phospholipid hydroperoxide glutathione peroxidase,PHGPx)的变化特点及过氧化损伤对 Ca2 + - Mg2 + - ATP酶活性的影响。方法 :测定了 2 9例 AMI患者红细胞 PHGPx、共轭双烯 (conjugated diene,CD)含量及 Ca2 + - Mg2 + -ATP酶活性。 2 0例健康同龄人做为对照。 PHGPx以 U rsini法测定 ;CD以 pryor法测定 ;Ca2 + - Mg2 + - ATP酶以 Hanahan法测定。结果 :与对照组相比 ,AMI患者红细胞 PHGPx、Ca2 + - Mg2 + - ATP酶水平明显降低 (P <0 .0 1)。结论 :AMI患者 PHGPx活性降低 ,抗磷脂过氧化能力低下。这可能是导致 AMI患者 Ca2 + - Mg2 + - ATP酶活性降低的原因之一。 Objective: Our aims were to observe the changes of erythrocyte phospholipid hydroperoxide glutathione peroxidase (PHGPx) in patients with acute myocardial infarction (AMI) and its effects on level of conjugated diene (CD) and Ca 2+ Mg 2+ ATPase activity. Methods: We measured contents of erythrocyte PHGPx and CD and activity of Ca 2+ Mg 2+ ATPase in 29 AMI patients and 20 controls, PHGPx with Ursini method, CD with Pryor method, and Ca 2+ Mg 2+ ATPase with Hanahan method. Results: The levels of erythrocyte PHGPx and Ca 2+ Mg 2+ ATPase were significantly lower in the AMI group than those in the control group (8.22±0.89) compared with (4.31±1.11) nmol NADPH/min·mgHb; (34.21±4.04) with (24.34±3.46) μmol protein gHb -1 ·h -1 20℃,P<0.01). The level of CD was significantly higher in patients than in the control group (0.64±0.08) with (1.25±0.22) nmol/mg Hb, P<0.01). There was significantly negative relationship between the levels of PHGPx and CD in AMI patients (r=-0.53, P<0.01). Conclusion: The decrease of PHGPx level in AMI patients may increase phospholipid peroxidation. It may be one of the causes of lower Ca 2+ Mg 2+ ATPase activity.
