摘要
目的探讨细胞周期素D1(Cyclin D1)基因多态性与大肠癌遗传易感性的相关性。方法采用聚合酶链反应-限制性片段长度多态性(PCR—RFLP)方法,检测125例大肠癌与118例健康对照者Cyclin D1基因第870位核苷酸A/G(A870G)的多态性。结果两组Cyclin D1基因型频率差异有统计学意义(P〈0.05)。与AG及GG基因型携带者之和相比,AA基因型携带者患大肠癌的相对风险度(OR)为2.41,95%可信区间为(1.39,4.17)。根据不同的临床特征分析,AA基因型频率在不同年龄或不同性别的结肠癌患者中,差异均无统计学意义(P〉0.05),但在远侧结肠癌[OR:2.58,95%CI(1.22,5.45),P=0.012]、低分化[OR=2.24,95%CI(1.07,4.70),P:0.031]和DuckesC期[OR=2.25,95%CI(1.08,4.69),P=0.029]患者中明显升高。结论Cyclin D1基因多态性与大肠癌及与肿瘤部位、分化程度和Dukes分期显著相关。
Objective To explore the relationship between gene polymorphism of Cyclin D1 and susceptibility to colorectal cancer. Methods Gene polymorphism of Cyclin D1 (A870G)was detected among 125 patients with colorectal cancer and 118 healthy controls by polymerase chain reaction- restriction fragment length polymorphism (PCR- RFLP). Results There was significant difference in Cyclin D1 genotype frequency between patients with colorectal cancer and healthy controls ( P 〈 0.05 ). Odds ratio (OR) of AA genotype carriers was 2.41 with 95 % confidence interval (1.39,4.17) compared with A/G and GG genotype carriers. According to different clinical characteristics, there was no significant difference in AA genotype frequency between patients with colorectal cancer in different ages and gender(P 〉 0.05 ). However, it increased in patients with distal [ OR = 2.58,95 % CI ( 1.22,5.45 ), P = 0.012 ], poor differentiated [ OR = 2.24,95% CI ( 1.07,4.70), P = 0. 031 ] and Dukes C stage [ OR = 2.25,95% CI ( 1.08,4.69), P = 0.029 ] colorectal cancer. Conclusion Gene polymorphism of Cyclin D1 is related to genetic susceptibility to colorectal cancer, tumor location,differentiation as well as Dukes stage.
