芪参健骨颗粒对激素性股骨头坏死大鼠骨组织的影响 被引量:7

The Effect of Qishenjiangu Granules on the Bone Tissue of Steroid-induced Avascular Necrosis of Femoral Head in Rats

摘要 目的:明确芪参健骨颗粒对激素合并内毒素所致的股骨头坏死模型大鼠骨组织的影响,并探讨其作用机制。方法:SD大鼠随机分为空白组、模型组、低、中、高剂量给药组、阳性对照组6组,除正常组外其余各组均用激素加内毒素诱导建立股骨头坏死模型,造模与给药同时进行,低、中、高剂量组分别给予芪参健骨流浸膏15、30、60g·kg-1,阳性对照组给予丹郁骨康丸3g·kg-1,正常组、模型组给予等量生理盐水,给药8周后,腹主静脉取血测血钙血磷含量;取两侧股骨,左侧用于制作组织病理切片,右侧进行骨密度测定,三点弯曲试验。结果:芪参健骨颗粒低、中、高剂量组及阳性对照组骨密度、最大载荷量、单位面积最大载荷量、弹性模量及血钙、血磷含量均显著高于模型组;模型组骨密度、最大载荷量、单位面积载荷量、弹性模量、血钙血磷含量显著低于正常组。结论:芪参健骨颗粒具有防治股骨头坏死的作用,可能通过提高骨密度、骨强度及血钙血磷含量发挥作用。 Objective:To find out the effect of Qishenjiangu Granules on the bone tissue of rat models of steroid-induced avascular necrosis of femoral head (SINFH) by combining hormone with endotoxin, and study the underlying mechanisms. Methods: SD rats were randomly divided into the control group, the model group, the low, medium, high-dose group, and the positive control group, respectively. Each group was replicated SINFH experimental animal model with the exception of the control group. Low, medium, high-dose Qishenjiangu extractum liquidum 15, 30, 60g. kg-1 were administered respec- tively in the three treatment groups, 3g. kg~ Danyugukang pills in the positive control group, and equal amount of normal saline both in the control group and the model group. After 8-week intragastric pre-administration, rats were sacrificed for blood samples from abdominal vein to determine the contents of calcium and phosphorus. Femurs from both sides were taken, the left side for pathological sections, and the right side for determination of the bone density and three-point ben- ding test. Results:The bone density, the maximum loading, maximum loading per unit area and elasticity loading as well as the content of blood calcium and phosphorus of rats in the treatment groups and the positive control group were higher than those in the model group. The above indicators were much lower in model group than in control group. Conclusion:Qishen-- jiangu Granules can prevent and cure the SINFH probably by affecting bone density, bone strength and contents of calcium and phosphorus.
