摘要
目的:观察灌胃凤凰衣水解物对膝骨关节炎大鼠膝关节的影响,进而考察凤凰衣用于防治OA的合理性及可行性。方法:灌胃凤凰衣水解物及奥泰灵胶囊干预膝骨关节炎大鼠,观察软骨形态及软骨中Collagen-Ⅱ、MMP-3及蛋白多糖的指标变化,以及血清和关节液中IL-6,IL-1β,TNF-α的指标变化。结果:1.各用药组对降低膝骨关节炎大鼠血清IL-1β、IL-6、TNF-α表达,关节液中TNF-α的表达以及软骨中MMP-3表达,升高Collagen-Ⅱ的表达看,两用药组未见明显差异(P>0.05),但均优于模型组(P<0.01);2.在提高蛋白多糖含量及降低关节液中IL-6的表达方面,灌胃凤凰衣组优于灌胃奥泰灵组(P<0.01);3.在降低关节液中IL-1β的表达方面,灌胃奥泰灵组优于灌胃凤凰衣组(P<0.01)。结论:1.灌胃凤凰衣水解物能降低膝骨关节炎大鼠的炎症因子表达,提高关节软骨中蛋白多糖和Collagen-Ⅱ含量;2.凤凰衣中含有丰富的可以合成软骨基质中蛋白多糖的前体物质,是其改善OA指标的机制之一。
Objective:To observe the influence of intragastric administration with hydrolyzed eggshell membrane on knee os- teoarthritis in rats. Methods:Rats with knee osteoarthritis were intragastrieally administered hydrolyzed eggshell membrane or glucosamine hydrochloride. Cartilage shape, proteoglycan, collagen-Ⅱ and MMP-3 in cartilage were observed. The levels of IL-6, IL-1β and TNF-a in serum and joint fluid were measured. Results: 1. The serum levels of IL-1β, IL-6 and TNF-α, synovial fluid TNF-a level as well as MMP-3 in cartilage were decreased in both treatment groups, with no signifi- cant difference in collagen-Ⅱ (P〈0.05). 2. Proteoglyean in hydrolyzed eggshell membrane group was increased much more than that in glucosamine hydrochloride group. 3. IL-1β in synovial fluid in glucosamine hydrochloride group was de- creased much better than that in hydrolyzed eggshell membrane group (P〈0.01). Conclusion: 1. Hydrolyzed eggshell membrane can reduce inflammatory factor expression of rats with knee osteoarthritis and increase proteoglycan and Colla- gen-Ⅱ content in articular cartilage. 2. Eggshell membrane contains abundant precusor substance that forms proteoglycan in cartilage matrix, which is one of the mechanisms in improvement of osteoarthritis by eggshell.
