固本培元化瘀颗粒对紫癜性肾炎的实验研究

The Empirical Study on the Effects of Gubenpeiyuan Granules on Purpura Nephritis

目的:探讨固本培元化瘀颗粒对紫癜性肾炎小鼠模型的药效作用。方法:将110只昆明种小鼠随机分为2组,即空白对照组(20只)和模型制作组(90只)。采用脂多糖(LPS)+四氯化碳(CCl4)+牛血清白蛋白(BSA)联合给药6周的方法,模拟紫癜性肾炎进行造模。造模结束后,从两组分别随机选取5只小鼠,在光学显微镜下观察肾组织的形态变化,并结合尿中红细胞及蛋白的情况,确定造模成功小鼠。将模型制作成功的60只小鼠,再按体重随机分为4组:模型组,甲氰咪胍(60mg/kg)组,固本培元化瘀颗粒(50g/kg)、(100g/kg)剂量组。各组连续给药5周后检测肾小球系膜细胞数、肾小球系膜基质指数等指标。结果:固本培元化瘀颗粒各组与甲氰咪胍组的肾小球肿胀、充血程度及肾小球系膜区的宽度明显小于模型组(P<0.05)。与模型组比较,固本培元化瘀颗粒(50g/kg)、(100g/kg)剂量组小鼠肾小球系膜细胞数明显减少(P<0.05)。结论:固本培元化瘀颗粒可以改善或修复实验小鼠的肾脏病理损害,有效延缓肾小球硬化的进程。

objective：To investigate the effects of Gubenpeiyuan Granules （GBPY） on Henoch-Schonlein purpura nephritis. Methods：llO Kunming mice were randomly divided into 2 groups, namely control group （20 rats） and model group （90 rats）. The LPS,CC14 and BSA method combined with 6 weeks of on simulation Henoch-Schonlein purpura nephritis modeling. After the modeling, the two groups were ran- domly selected from 5 mice, to observe the morphological changes of renal tissues under the light microscope, and a combination of the red blood cell and protein in urine, to determine the successful model of mice. The model was made of 60 mice successfully, randomly divided into 4 groups： model group, tagamet group（6Omg/kg）, GBPY （50g/kg）, （100g/kg） dose group. Groups were administered continuously for 5 weeks after the pathological changes of renal tissue were observed, and the detection of glomerular mesangial matrix index. Result：The glomerular swelling, hyperemia and glomerular mesangial area width of GBPY group and tagamet group were significantly lower than that of model group（P〈0.05）. Compared with the model group, Scr, BUN of GBPY（50g/kg）, （100g/kg） dose group is decreased （P〈0.05）. Con- clusion： GBPY can improve or repair of renal damage in mice, reduce the mice serum creatinine and urea nitrogen index effectively to delay the progress of glomerular sclerosis.
