摘要
目的:研究不同强度的体外高压电场对肺癌细胞系A549细胞的黏附、迁移及侵袭等能力的影响。方法:选择处于生长周期的A549细胞,实验组共分为7个亚组,分别施加500~2 500V/cm的高压电场,对照组未施加电场处理。经生长抑制实验、黏附实验、侵袭及转移实验检验不同强度高压电场对A549细胞的影响。结果:1)细胞黏附能力:实验组各亚组的黏附细胞数明显少于对照组,实验组各亚组与对照组、实验组各亚组之间差异有统计学意义,P<0.001。2)细胞迁移能力:电场强度为500V/cm的实验亚组与对照组比较,差异无统计学意义,P=0.089;电场强度≥750V/cm的各实验亚组之间、及其与对照组比较,差异有统计学意义,P<0.001。3)细胞侵袭能力:电场强度为500和750V/cm的实验亚组与对照组比较,差异无统计学意义,P=0.181;电场强度≥1 000V/cm的实验亚组与对照组间比较,差异有统计学意义,P<0.001;电场强度为1 000~1 250V/cm的实验亚组和1 500~2 000V/cm实验亚组间比较,差异有统计学意义,P<0.001。结论:体外高压电场可以明显抑制A549细胞的黏附、迁移和侵袭等能力;随着电场强度的增加,肿瘤细胞的抑制程度也随之增加。
OBJECTIVE:To evaluate on A549 lung carcinoma cells' ability of adhesion,migration and invasion in- duced by different high voltage electric field in vitro. METHODS:Selecting A549 lung carcinoma cells being in growth cy- cle were selected. There were seven experimental groups, and exert high voltage electric field from 500 V/cm to 2 000 V/cm. In addition,the control group did not exert high voltage electric field. The effects of different high voltage e- lectric field on A549 lung carcinoma cells were inspected by growth inhibition test,adhesion test,invasion test and migra- tion test. RESULTS: 1) Cell adhesion ability:The number of adherent cells for each experimental subgroup was significant- ly less than that of the control group (P^0. 001). 2) Cell migration ability: No significant differences were found between the 500 V/em experimental subgroup and the control group (P:0. 089). Significant difference was found among all the experimental subgroups and the control group when the electric field strength was increased upon 750 V/cm (P^0. 001). 3)Cell invasion ability: No significant differences were found among the 500V/cm and 750V/cm in experimental subgroup and the control group (P=0. 181). Significant difference was found among all the experimental subgroups and the control groups when the electric field strength was increased upon 1 000 V/cm (P^0. 001). Significant difference was found be- tween the 1 000--1 250 V/cm and 1 500--2 000 V/cm in experimental subgroups (P^0. 001). CONCLUSIONS: High- voltage electric field can inhibit A549 lung carcinoma cells' ability of adhesion,migration and invasion significantly. With the electric field intensity increases, the degree of inhibition of tumor ceils also increases.
