摘要
目的:探讨结直肠癌组织中Tip60蛋白的表达及其临床意义。方法:选择2010-02-01-2011-02-01嘉兴市第一医院行手术治疗的结直肠癌患者100例和结直肠腺瘤患者20例,采用免疫组织化学检测Tip60蛋白在100例结直肠癌组织和相应癌旁正常组织(距离癌组织>3cm)及20例结直肠腺瘤组织中的表达及差异,观察Tip60和Ki-67及Bcl-2蛋白在结直肠癌组织中表达的相关性。结果:Tip60蛋白在结直肠癌组织中的阳性表达率为24%(24/100),癌旁正常组织为72%(72/100),结直肠腺瘤组织为75%(15/20),结直肠癌中的表达明显低于癌旁组织,χ2=46.154,P<0.001;也低于腺瘤组织,χ2=19.761,P<0.001;癌旁组织及腺瘤组织的表达差异无统计学意义,χ2=0.075,P=0.781。Tip60的表达与患者性别(χ2=1.629,P=0.202)、年龄(χ2=1.170,P=0.279)、肿瘤浸润深度(χ2=3.102,P=0.078)及淋巴结转移(χ2=0.718,P=0.397)无关,而与肿瘤大小(χ2=14.649,P<0.001)、肿瘤分化程度(χ2=6.469,P=0.039)和Duke分期(χ2=4.702,P=0.030)相关。Tip60和Ki-67的表达呈负相关,r=-0.273,P=0.006,Tip60和Bcl-2蛋白的表达呈负相关,r=-0.216,P=0.031。Logistic回归分析发现,肿瘤大小是结直肠癌Tip60蛋白表达的独立相关因素,B=-1.833,P<0.001。结论:Tip60蛋白在结直肠癌组织中表达下调,且与结直肠癌的进展有关,Tip60可作为预测结直肠癌进展和预后的生物学指标。
OBJECTIVE: To investigate the expression of Tip60 protein in colorectal cancer tissue and its clinical sig- nificance. METHODS: Immunohistoehemical technique was adopted to detect the protein expression level of Tip60 protein in 100 cases of colorectal cancer,adjacent tumor tissues (3 era off the turner region), and 20 eolorectal adenoma tissues, meanwhile the relationship between the expression of Tip60 and the expression of Ki-67 ,Bcl-2 protein were analyzed. RE- SULTS: The positive rate of Tip60 in eolorectal cancers was 24% X24/100) and in adjacent tumor tissues and in adenoma tissues the positive rate were 72% (72/100) ,75% (15/20). The positive rate of Tip60 in coloreetal cancer was significant- ly lower than that of adjacent tumor tissues(x2 =46. 154,P=0. 001) and adenoma tissues (X2 =19. 761,P=0 001). The expression of Tip60 was of no statistical difference in adjacent tumor tissues and colorectal adenoma tissues X2 =0. 075, P=0. 781). No significant correlations were observed between the expression of Tip60 in colorectal cancer and patient gender X2 = 1.629, P = 0.202 ), age ( X2 = 1.170, P = 0.279 ), depth of tumor invasion ( 2 = 3. 102, P : 0.078 ), or lymph node rnetastasis(X2= 0. 718, P : 0. 397). Tip60 down-regulation showed significant correlation with larger tumor size ( X2 : 14. 649, P~ 0. 001 ), poorly differentiated type ( X2 : 6. 469, P ~ 0. 039 ) and higher Duke ' s stage ( 2 = 4. 702, P : 0. 030). Tip60 protein expression in colorectal cancer was negatively correlated with Ki-67 (r= --O. 273, P : 0. 006) and Bcl-2 protein expression in colorectal cancer (r=--0. 216 ,P=O. 031). Logistic regression analysis showed that the tumor size was independent correlation factor of Tip60 expression in colorectal cancer tissues (B:- 1. 833, P=0 001). CON- CLUSION: These results suggest that Tip60 is more frequently down-regulated in advanced colorectal carcinoma, Tip60 may serve as a new parameter for the prognostic prediction of colorectal cancer.
