摘要
设计并合成了两段各25bp长的寡聚核苷酸片段,末端标记后与成年奶牛染色体组DNA杂交。结果表明,它们只与公牛染色体DNA发生阳性杂交。以这两个寡聚核苷酸片段作为引物对奶牛染色体组DNA进行PCR扩增,在模板为公牛染色体DNA时才能扩增到一条约180bp左右的DNA片段。两种实验结果表明:这两段寡聚核苷酸片段具有牛雄性特异性,因而可用以早期鉴定牛胚胎性别。
Since the sex chromosoms of the male and female cattles are XY and XX respectively, a Y-specific (or male specific) nucleic acid probe to identify the sex of cattle embryos could be used. Two polynucleotide fragments were synthesized to be 25-bp in length, and they showed male-specific character in the dot-blotting hybridization and PCR experiments. This method could be used for the identification of the sex of cattle preimpantation embryos.
