摘要
目的观察白藜芦醇对高脂饲料诱导的肥胖易感和肥胖抵抗大鼠体重的影响并探讨其相关的分子机制。方法 58只6周龄雄性SD大鼠,分别给予高脂饲料和基础饲料8w,根据对照组的平均体重从高脂组大鼠筛选出肥胖易感(OP)和肥胖抵抗(OR)大鼠,再分别将OP和OR组大鼠随机分为两组,一组继续高脂喂养(OP/OR),另一组在高脂饲料中的同时添加250 mg/(kg bw·d)的白藜芦醇(OP-R/OR-R),对照组继续饲喂基础饲料(C)。干预结束后,称取大鼠的体重和内脏脂肪重量,分离血清测定血糖血脂和胰岛素含量,脂肪组织病理切片观察脂肪蓄积情况,实时定量PCR检测脂肪组织AMPKα和Sirt1的基因表达。结果白藜芦醇干预后,与未干预组相比,OP-R组的体重、体脂含量和能量利用率显著降低;血清中总胆固醇(TC)、甘油三酯(TG)、胰岛素(FINS)含量和胰岛素抵抗指数(HOMA-IR)也显著下降,高密度脂蛋白胆固醇(HDL-C)水平显著增加(P<0.05);脂肪细胞体积变小,脂肪蓄积量减少;脂肪组织AMPKα1、AMPKα2和Sitr1的mRNA表达显著增加(P<0.05);而OR-R组则无显著变化。结论白藜芦醇通过促进肥胖易感大鼠脂肪组织AMPKα1、AMPKα2和Sitr1的基因表达降低高脂膳食诱导肥胖易感大鼠的体重和脂肪蓄积量,而对肥胖抵抗大鼠的体重则无显著影响。
Obejective To investigate the different effect of resveratrol on the body weight of high-fat diet induced obesity prone and resistant rats and the related mechanism. Methods Male Sprague-Dawley rats were randomly divided into high-fat group (HF, n=48) and chow food group(C, n=10), Rats in high-fat group were subdivided into obesity obesity prone (OP) and resistance (OR) group according to the final body weight compared with the C group after 8 w. One half of the OP and OR rats were kept on high fat diet (OP/OR) and the other half were switched to high fat diet with resveratrol (250 mg/(kg-bw.d)) (OP-R/OR-R) for the following 12 w. Body weight, visceral fat pads, energy efficiency were measured. Serum lipids, glucose and insulin were assayed. AMPKct and Sirtl mRNA expression were determined by real time polymerase chain reaction (RT-PCR). Results After 12 w resveratrol intervention, the body weight, body fat , energy efficiency, TC, TG, FINS and HOMA-IR in OP-R group were reduced compared with OP group, HDL-C level was increased, as well as the mRNA expressions of AMPKctl, AMPKct2 and Sirtl in white adipose tissue were significantly increased (P〈0.05). There was no difference between OR-R and OR group. Conclusion Resveratrol reduces weight gain induced by high fat diet in OP but not OR rats, which may be associated with the increased mRNA expression of AMPKctl, AMPKct2 and Sirtl in white adipose tissue.
