硫氢化钠对慢性脑缺血大鼠学习记忆及TH表达的影响

摘要 目的:探讨NaHS对慢性脑缺血大鼠学习记忆及TH表达的影响。方法采用Sarti等改良的分次结扎双侧颈总动脉术制作慢性脑缺血大鼠模型,术后8周五组(假手术组,慢性脑缺血组,低剂量组,中剂量组,高剂量组)大鼠Morris水迷宫测试后断头取脑检测H2S的含量,免疫组化检测TH的表达。结果:水迷宫实验结果:慢性脑缺血组学习记忆能力显著下降,与假手术组比较有显著性差异(P〈0.05),NaHS中、高剂量组学习记忆能力显著提高,与慢性脑缺血组比较有显著性差异(P〈0.05),说明NaHS能有效改善大鼠的学习记忆功能。慢性脑缺血组大鼠脑内H2S含量明显降低,与假手术组比较有显著性差异(P〈0.05);NaHS中、高剂量组H2s含量明显升高,与慢性脑缺血组比较有显著性差异(P〈0.05)。大鼠TH的表达水平慢性脑缺血组明显升高,与假手术组比较有显著性差异(P〈0.05),硫氢化钠中剂量组、高剂量组与慢性脑缺血组比较有显著性差异(P〈0.01)。结论NaHS可明显增强慢性脑缺血大鼠脑组织H2S生成,提示H2S可能改善慢性脑缺血大鼠学习记忆功能,其机制可能与上调TH的表达有关。 Objictive:To investigate Effects of sodium hydrosulfide on learning and memory abilities and the expression of TH in chronic cerebral ischemic rats. Methods:Sarti's 2VO was used to make the chronic cerebral ischemic rats. All rats were randomly divided into sham operation group, chronic cerebral is- chemia group,low dose group, middle dose groupand high dose group. Morris water maze was used to evaluate the effects of sodium hydrosulfide on chronic cerebral isehemic rats in their learning and memory abilities; The SPSS16.0 Statistics software was used to analyze data of variance. Results~ 1. Water maze test results. The rats learning and memory ability in chronic cerebral ischemia group significantly decreased,and which was lower than that in the sham group (P〈0.05). The rats' learning and memory ability of the middle and high-dose group of NariS significantly increased,2 The content of H2S in brain in the chronic cerebral ischemia group was significantly lower than that in the sham group (P〈0.05) . The content high-dose group of NariS was significantly high er than that in the chronic cerebral ischemia group (P〈0.05). 3 The level of TH expression in rats brain in chronic cerebral ischemia and which was higher than that in the sham group(P〈0.05) ,The level of TH expression in middle and high dose group of sodium was higher than that in chronic cerebral ischemia (P〈0.01). Conclusion: Naris can significantly enhance the content of H2S in rat brain tissue in the chronic cerebral ischemia,suggesting that H2S may im- prove ability in rats with chronic isehemia,the mechanism may be related to increased the level of TH expression.
