Genetic Diversity and Genetic Differentiation among Different Geographic Populations of Oedaleus infernalis Saussure(Orthoptera:Acridoidea)

摘要 应用线粒体细胞色素b(Cytb)基因序列对10个地理种群黄胫小车蝗(Oedaleus infernalis Saussure)的遗传多样性及遗传分化进行了初步研究。结果表明:在获得的168 条序列样本中,共发现了14 个变异位点,定义了15 种单倍型。总单倍型多样性指数Hd 为0.462,种群内单倍型多样度在0.125-0.625 范围之内。总群体的Fst 为0.0812,Gst 为0.0991,Nm为4.55,表明黄胫小车蝗各地理种群之间存在着较为充分的基因交流。分子方差分析(AMOVA)结果表明,黄胫小车蝗的遗传差异主要发生在种群内部,占方差比率的91.53%。单倍型中介网络图及系统树显示,各种群中的单倍型分布混杂,没有形成明显的地理格局。 The mitochondrial Cytb gene was used to investigate genetic diversity and genetic differentiation among 10 geographic populations of Oedaleus infernalis Saussure. In the total 168 tested sequences, 14 variable sites and 15 haplotypes were observed. The mean haplotype diversity of all populations was 0.462 and the value within populations ranged from 0.125 to 0.625. The overall Fst was 0.0812, Gst was 0.0991 and Nm was 4.55. These results suggested that gene flow between different geographic populations was sufficient. The results of AMOVA suggested that the genetic variation mainly occurred within populations (91.53%). The median-joining network and phylogenetic tree of haplotypes revealed no obvious geographical pattern for the haplotypes distributed in mixed structure.
