Chemical constituents from root tubers of Rehmannia glutinosa

摘要 目的研究地黄Rehmanniaglutinosa块根的化学成分。方法通过正相硅胶、SephadexLH-20以及反相HPLC柱色谱方法对地黄的化学成分进行分离纯化,应用谱学技术鉴定化合物的结构。结果从地黄块根乙醇提取物中分离得到29个化合物,分别鉴定为1,2,5,6-四氢-1-甲基-2-氧代-4-吡啶乙酸(1)、5-deoxyantirrhinoside(2)、5-deoxylamiol(3)、去乙酰野芝麻苷(4)、栀子新苷(5)、genipin-gentiobioside(6)、genamesideC(7)、68-hydroxy-2-oxabicyclo[4.3.0]△^8-9_nonen-l-one(8)、焦地黄素D(9)、焦地黄素E(10)、地黄苷(11)、sculponiside(12)、2-phenylethyl—O-β-9-xylopymnosyl-(1—6)-β-D—glucopyranoside(13)、毛蕊花糖苷(14)、红景天昔(15)、leueoseeptosideA(16)、焦地黄苯乙醇苷D(17)、deacyl-martynoside(18)、焦地黄苯乙醇苷A1(19)、焦地黄苯乙醇苷B1(20)、3,4-dihydroxy—p-phenethyl—O-α-L-rhamnopyranosyl-(1→3)-O-β-D—galacopyranosyl-(1→6)-4-O—caffeoyl-β-D—glucopyranoside(21)、1-(4-methy-2-furanyl)-2-(5-methyl-5-ethenyl-2-tetrahydrofuranyl)-propan-1-one(22)、2-methoxy-4-methylphenyl-O-β-D—apiofuranosyl-(1→6)-β-D-glueopyranoside(23)、rhamnopyranosylvanilloyl(24)、syringicacid-4·O-α-L-rhamnopyranoside(25)、(7R,8S,7'R,8S,7'4,9,4’,9'-tetrahydroxy-3,3'-dimethoxy-7,7'-epoxylignan9-O-β—D-glucopyranoside(26)、1-methyl-1,2,3,4-tetrahydro-β—carboline-3-carboxylicacid(27)、异直蒴苔苷(28)、直蒴苔苷(29)。结论29个化合物包括9个环烯醚萜类(2-10)、11个苯乙醇苷类(11-21)和9个其他类型(1、22-29)化合物,其中化合物1为新化合物,命名为地黄啶,化合物2~8和24-29为首次从该属植物中分离得到。经体外活性筛选发现化合物1、4、13、15、24、25、27和28对D-半乳糖胺诱导的人肝细胞HL-7702损伤有较明显的保护作用。 Objective To investigate the chemical constituents in the root tubers ofRehmannia glutinosa. Methods The compounds were isolated from the ethanol extract in the root tubers of R. glutinosa by a combination of various chromatographic techniques including column chromatography over silica gel, Sephadex LH-20 columns, and reversed-phase HPLC. The structures were identified by spectroscopic analysis including MS and NMR data. Results Twenty-nine compounds were isolated and identified as 1, 2, 5, 6-tetrahydro-l-methyl-2-oxo-4-pyridineacetic acid (1), 5-deoxyantirrhinoside (2), 5-deoxylamiol (3), lamiol (4), gardoside (5), genipin-gentiobioside (6), genameside C (7), 6β-hydroxy-2-oxabicyclo [4.3.0] A8-9-nonen-l-one (8), jioglutin D (9), jioglutin E (10), martynoside (11), sculponiside (12), 2-phenylethyl-O-β-D-xylopyranosyl-(1→6)-β-D-glucopyranoside (13), acteoside (14), salidroside (15), leucosceptoside A (16), jionoside D (17), deacyl-martynoside (18), jionoside A1 (19), jionoside Bl (20), 3, 4-dihydroxy-β-phenethyl-O-α-L-rhamnopyranosyl( 1→ 3 )-O-β-D-gaoacopyranosyl-( 1→6)-4-O-caffeoyl-β-D-glucopyranoside (21), 1-(4-methy-2-furanyl)-2-(5-methyl-5-ethenyl-2-tetrahydrofuranyl)-propan-l-one (22), 2-methoxy-4-methylphenyl-O-β-D- apiofuranosyl-(1→6)-β-D-glucopyranoside (23), rhamnopyranosyl vanilloyl (24), syringicacid-4-O-ct-L-rhamnopyranoside (25), (TR, 8S, 7'R, 8'S)-4, 9, 4', 9'-tetrahydroxy-3, Y-dimethoxy-7, 7'-epoxylignan 9-O-15-D-glucopyranoside (26), 1-methyl-1, 2, 3, 4-tetrahydro13-carboline-3-carboxylic acid (27), tachioside (28), and isotachioside (29). Conclusion Among the isolated 29 compounds, there are 9 iridoid and iridoid glycosides (2-10), 11 phenethylalcohol glycosides (11-21), and 9 other components (1 and 22-29). Compound 1 is a new compound named rehmannidine, and compounds 24 and 24-29 are isolated from the plants ofRehmannia Libosch. ex Fisch. et Mey. for the first time. In the preliminary assays, compounds 1, 4, 13, 15, 24, 25, 27, and 28 exhibit the moderate hepatoprotective activites against D-galactosamine-induced HL-7702 cell damage.
