钙蛋白酶抑制蛋白(CAST)基因是肉质候选基因,采用聚合酶链式反应-单链构象多态(PCR-SSCP)方法对59只小尾寒羊和51只巴寒杂交一代的CAST基因第6外显子进行多态性检测,研究其多态性与肉质的相关性。结果表明:在CAST基因第6外显子上检测到3种基因型,AA、AB和AC型,在小尾寒羊中,其基因型频率分别为0.42、0.37和0.41。A、B、C的基因频率分别为0.71、0.19和0.10。多态信息含量(PIC)为0.40。在巴寒杂交群体中,其基因型频率分别为0.41、0.53和0.06。A、B、C的基因频率分别为0.71、0.26和0.03。PIC为0.36。在小尾寒羊背最长肌处,AB型与AA型的嫩度存在显著差异,AB型嫩度优于AA型。在臂三头肌处,AA、AB型的红度值存在显著差异,AA型高于AB型。在股二头肌处,AA、AB型的黄度值存在显著差异,AA型高于AB型。在巴寒杂交一代群体中背最长肌处,AB、AC型的黄度值存在显著差异,AB型高于AC型;AB、AC型的亮度值存在显著差异,AC型高于AA型和AB型。
Calpastatin(CAST) gene is an candidate gene for meat quality. Polymerase chain reactionsingle strand conformation polymorphism(PCR-SSCP) method was used to determine the polymorphism of exon 6 of CAST gene in 59 Small fat-tail sheep and 51 Bamei×Small fat-tail sheep F1, the correlation between CAST gene and meat quality traits were analyzed. Three genotypes were identified as AA, AB and AC. The genotype frequencies in Small-tailed sheep were 0.42, 0.37 and 0.41. Frequencies of A, B, C were 0.71, 0.19 and 0.10. Polymorphism information content was 0.40. The genotype frequencies in Bamei×Small fattail sheep F1 were 0.41, 0.53 and 0.06. Frequencies of A, B, C were 0.71, 0.26 and 0.03. Polymorphism information content was 0.36. In Small fat-tail sheep, the tenderness of AB genotype was significant higher than AA genotype in longissimus dorsi, red/green colours(a*) of AA genotype was significant higher than AB genotype in triceps, yellow/blue colours(b*) of AA genotype was significant higher than AB genotype in biceps femoris. In Bamei×Small fat-tail sheep F1, yellow/blue colours(b*)of AB genotype was significant higher than AC genotype and lightness(L*) of AC genotype was significant higher than AB genotype in longissimus dorsi.
