摘要
UBE2C(Ubiquitin-conjugating enzyme 2C)属于E2泛素偶联酶家族成员,较为明确的作用是调控有丝分裂检验点以及控制细胞周期进程,但其在小鼠胚胎中的作用尚不明确。本文首先探讨了小鼠多种组织以及胚胎不同发育时期UBE2C表达图谱,其次在小鼠受精卵中通过显微注射过表达UBE2C分析其对小鼠早期胚胎发育的影响。结果表明:过表达UBE2C后导致胚胎2-细胞发育阻滞,证明UBE2C在胚胎发育过程中具有重要功能,其作用可能是通过降解卵母细胞储存的母源蛋白,从而确保小鼠早期胚胎正常发育进程。
UBE2C( Ubiquitin- conjugating enzyme 2C )belongs to the E2 family of ubiquitin conjugating enzymes. It regulates checkpoint during mitosis and cell cycle progression. However, the functions are not known during mouse pre -implantation development. In this study, we showed that UBE2C has significantly higher expression level in ovaries and pancreas than in other tissues. Further analysis of pre -implantation embryos, we found that UBE2C has relative high level expression in morula. Overexpression of UBE2C in zygote using microinjection led to 2 - cell arrest during pre - implantation development. We propose that UBE2C functions in degradation of maternal proteins, which could affect development of pre - implantation embryo.
