摘要
应用免疫荧光技术以生物素化的荆豆素 - ( UEA- )为探针 ,检测其结合位点 α-L-岩藻糖在小鼠妊娠 1~ 8d、假孕 1~ 6 d及发情周期子宫中的表达情况 ,并以摘除卵巢的小鼠为模型观察雌激素、孕酮对小鼠子宫α-L-岩藻糖表达的影响。结果 ,UEA- 的结合位点主要位于妊娠 1~ 4 d的子宫腔上皮及腺上皮中。在假孕小鼠 ,UEA- 的结合位点主要位于假孕 1~ 2 d的子宫腔上皮及腺上皮中 ,假孕 3~ 4 d时在这些部位的结合逐渐减少或消失。另外 ,在发情周期各期的子宫腔上皮和腺上皮中也发现很多 UEA- 的结合部位 ,在发情期明显增多 ,当用雌激素处理时在这些部位的结合明显增加。这些表明 ,UEA- 在子宫腔上皮和腺上皮上的结合位点可能与胚胎的存在与否无关 ,而是受母体雌激素的调节。但胚胎着床后 UEA- 的结合位点消失 ,以后又在初级蜕膜中出现 ,表明 UEA-
Ulex Europeus(UEA Ⅰ) was used as a probe to study the expression of α L Fucose in mouse uterus during early pregnancy,pseudopregnancy and estrous cycle.Ovariectomized mice were also used for the study on hormonal regulation of α L Fucose expression.The binding sites of UEA Ⅰ were detected both in luminal and glandular epithelia of mouse uterus on days 1 4 of early pregnancy.During pseudopregnancy,the level of UEA Ⅰ binding sites was high on days 1 and 2,and low on days 3 and 4.The binding sites of UEA Ⅰ were detected in luminal and glandular epithelia during the estrous cycle,with the highest level to be at estrus.UEA Ⅰ binding sites in luminal and glandular epithelia were obviously increased by estrogen.These results suggest that the binding sites of UEA Ⅰ in luminal and glandular epithelia of mouse uterus may be under the control of maternal hormones and not related to the presence of embryos.The binding sites of UEA Ⅰ may play a role in the decidualization since they were detected during decidualization and not detected following implantation.
