摘要
以香水百合的鳞片愈伤组织为试验对象进行生物反应器培养研究。结果表明:生物反应器在暗光条件下,气流量为20mL·min-1时更有利于百合鳞片愈伤组织的正常生长,最佳诱导和增殖培养基为含有BA0.5mg·L-1的MS培养基,最佳固体分化培养基为含有BA 0.5mg·L-1和NAA0.5mg·L-1的MS培养基。相对于固体培养而言,生物反应器增加了小仔球的分化能力;避免不适气流量引起的组织褐化是影响培养物最佳生长发育的关键因子之一。
Calli were induced from bulb scales of perfume lily,and then cultured in airlift bio-reactors.The best growth of calli was achieved with air supplement of 20mL·min-1 air-flow for these bioreactors in darkness.The optimum medium for both callus induction and bulking up was liquid MS medium supplemented with 0.5mg·L-1 BA.Bulblets differentiated and grew after callus tissue was transferred onto semi-solid MS medium containing 0.5mg·L-1 BA and 0.5mg·L-1 NAA.Compared with semi-solid culture,bulblet differentiation was enhanced significantly in airlift bioreactors.It was essential to avoid tissue browning by using optimum air-flow rates for the best growth and development of plant tissue in the bioreactor.
