不同能量水平对断奶獭兔皮肤组织IGF-1和IGF-1R基因表达的影响 被引量:5

Effects of Different Dietary Energy on IGF-1 and IGF-1R Gene Expressions in Skin Tissue of Weanling Rex Rabbits

摘要 旨在研究不同消化能水平的日粮对断奶獭兔皮肤组织中IGF-1和IGF-1R基因表达的影响。选取45日龄体质量(1 133±71g)的断奶獭兔160只,随机分为4个组,每组40个重复,每个重复1只,分别饲喂消化能为9.70MJ/kg(Ⅰ组)、10.50MJ/kg(Ⅱ组)、11.30MJ/kg(Ⅲ组)和12.10MJ/kg(Ⅳ组)的饲粮,预试期7d,然后进入正式期。在75日龄和105日龄时分别屠宰,取背中部约1cm2皮肤组织样,提取总RNA,反转录,荧光定量PCR法检测皮肤组织中与毛品质相关基因的相对表达水平。结果表明:在獭兔75日龄时,Ⅲ组IGF-1基因的表达量显著高于Ⅰ组(P<0.05),Ⅲ组IGF-1R基因的表达水平显著高于Ⅰ组(P<0.05)。在獭兔105日龄时,Ⅲ组IGF-1和IGF-1R基因的表达水平仍然最高,但各组间差异不显著。说明在獭兔断奶至约105日龄时,消化能水平维持在11.30MJ/kg较为合适,此时IGF-1和IGF-1R基因mRNA表达水平最高,在充足能量的保证下,獭兔皮肤毛囊与毛发能够快速稳定生长。 The experiment was conducted to research the effects of different dietary energy on gene ex- pressions ofIGF-1 andIGF-1R in skin tissue of weaning rex rabbits. One hundred and sixty 45d aged rex rabbits with an average body mass of (1 133±71) g were randomly divided into 4 groups with 40 replicates, and each replicate contained 1 rabbit. Rabbits in the 4 groups were fed experimental diets containing 9.70 MJ/kg (Group I ), 10.50 MJ/kg(GroupⅡ), 11.30 MJ/kg(GroupⅢ), 12.10 MJ/kg DE(Grouplg), respectively. The experiment included a pretrial period of 7 days and an experimental period of 60 days. At the age of 75 days and 105 days, 1 cm2 skin tissue was collected from central back of slaughtered rabbits respectively and used for total RNA extraction and reverse transcription as well as gene expression of hair quality related IGF-J and IGF-1R by real time PCR. The results showed that at the age of 75 days, the expressions ofIGF-1 gene was significantly higher in Ⅲgroup than in I group (P〈0.05) ; the expression ofIGF-1R gene was significantly higher in Ⅲ group than in group (P〈0.05), too. At the age of 105 days, the expressions of bothIGF-1 andIGF-1R genewere the highest in Ⅲ group, but the expressions of every group has no significant difference suits indicated that the expressions of IGF-1 and IGF-1R gene were the highest when the dig ergy level was 11.30 MJ/kg during the growth period of rex rabbits. And skin hair follicles could grow fast and steadily under the condition of sufficient energy supply. The re estive en and woolcould grow fast and steadily under the condition of sufficient energy supply.
