摘要
目的 观察γ干扰素 (IFNγ)在人体内供皮区创面上对成纤维细胞的生物学影响 ,探讨γ干扰素应用于临床防治瘢痕的理论依据。方法 用免疫组织化学方法检测 15例烧伤后患者的中厚皮片供皮区创面愈合过程中IFNγ对成纤维细胞产生Ⅰ、Ⅲ型胶原及向成纤维细胞分化等生物学作用的影响。结果 IFNγ处理过的创面肉芽组织中 ,Ⅰ、Ⅲ型胶原合成减少 ,α SMA的表达比例下降 (P〈0 .0 5 )。结论 IFNγ可以抑制创面成纤维细胞向肌成纤维细胞的分化以及合成胶原的功能 ,起到抑制纤维化的作用 。
Objective To observe the biological influence of Interferon Gamma (IFNγ) on the human wound fibroblasts, and explore a theoretical basis for the clinical application of IFNγin the prevention and treatment of wound surface.Methods Fifteen burned patients were selected for the research, and the immunohistochemistry was applied to examine the biological influence of the IFNγ on the type Ⅰ, type Ⅲ collagen and the differentiation of the fibroblasts in the process of healing at the site of split thickness skin graft.Results In the granulation tissue processed by IFNγ,type Ⅰ and type Ⅲ collagen decreased, and the SMA expression ratio was lowered( P <0.05).Conclusion IFNγcan downregulate the biological activities of fibroblasts in vivo, which inhibited collagen synthesis and myofibroblast differentiation. It is confirmed that IFN γcan be used for the prevention and treatment of hypertrophic scars.
