摘要
为了提高重组亚油酸异构酶的活力,试验对重组亚油酸异构酶诱导表达过程中的诱导时机、诱导时间、诱导剂浓度和诱导温度4个影响因素进行了研究,并进行了优化。正交试验表明,最佳诱导条件为:诱导时机为诱导前培养菌体7 h,诱导时间为10 h,诱导剂浓度为0.15 mM,诱导温度为30℃。最佳诱导条件下,诱导出的亚油酸异构酶的酶活力平均可达3 849.686 0 U/ml。结果显示,通过对诱导条件进行优化所得的亚油酸异构酶具有较高的酶活力。
In order to improve the activity of the recombinant linoleic acid isomerase in this experi- ment, four factors for the optimization of induced culture were studied, including induction timing, induced time, inducer concentration and inducing temperature. The nrthogonal experiment shows that, activity of linoleic acid isomerase is up to 3 849.686 0 U/ml, when the timing of induction is recombinant strain growth 7 h after, induction time was 10 h, inducer concentration of induction is 0.15 mM and induction temperature is 30℃. The results show that we can get the higher activity of linoleie acid isomerase by optimization of the induction conditions.
