摘要
目的建立同时测定高良姜药材中槲皮素、山柰素、乔松素、高良姜素共4种黄酮类成分含量的RP-HPLC法。方法采用Agilent ZORBAX SB-C18色谱柱(250mm×4.6mm,5μm),流动相为甲醇-乙腈-水-甲酸(体积比24:26:50:0.5),检测波长为280nm,流速为1.0mL·min^-1,柱温为30℃。结果槲皮素、山柰素、乔松素、高良姜素的线性范围分别为0.50~4.95mg·L^-1、0.53-5.28mg·L^-1、2.08-20.8mg·L^-1和10.29-102.9mg·L^-1,线性关系良好。平均加样回收率分别为98.3%、99.4%、98.6%和99.8%,RSD分别为1.6%、1.8%、1.6%和0.9%(n=9)。应用该方法测定了10批高良姜药材中4种黄酮类成分的含量。结论该方法准确、可靠,可为高良姜药材的质量控制提供参考依据。
Objective To develop an RP-HPLC method for simultaneous determination of quercetin, kaempferide, pinocembrin and galangin in Alpinia officinarum Hance. Methods Separation was carded out on an Agilent ZORBAX SB-C18 column (250 mm ×4. 6 mm,5 μm) with an isocratic mobile phase consisting of methanol, acetonotrile, water and formic acid ( V: V: V: V = 24 : 26 : 50 : 0. 5 ) at a flow rate of 1.0 mL. min ^-1. The detection wavelength was set at 280 nm and the column temperature was 30 ℃. Results The linear ranges of quercetin, kaempferide, pinocembrin and galangin were 0. 50- 4.95 mg· L ^- 1 , 0. 53 - 5.28 mg· L^- 1,2. 08 - 20. 8 mg·L ^-1 and 10. 29 - 102.9 mg· L^- 1, respectively. The average recoveries of quercetin,kaempferide, pinocembrin and galangin were 98.3%, 99.4%, 98.6% and 99. 8%, the relative standard deviations (RSDs) were 1.6% ,1.8% ,1.6% and 0. 9% (n =9). Conclusions The method is accurate and reliable, and can be used for the quality control of Alpinia officinarum Hance.
