摘要 目的建立同时测定葡萄籽提取物中原花青素B1和B2含量的方法。方法采用RP-HPLC法。色谱柱为Kromasil C18柱(250mm×4.6mm,5μm),以甲醇-体积分数0.1%磷酸溶液为流动相,梯度洗脱,检测波长280nin。结果原花青素B1和B2的质量浓度分别在23.0-230mg·L^-1(r=0.9991)和14.5~145mg·L^-1(r=0.9994)内与峰面积呈良好的线性关系;平均回收率分别为96.3%和97.5%,RSD分别为1.4%和1.3%。结论该方法简便、准确、重复性好,可用于测定葡萄籽提取物中原花青素B1和B2的含量。 Objective To develop an RP-HPLC method to determine the procyanidin B1 and B2 in grape seed extract simultaneously. Methods The sample was analyzed on Kromasil C18 column (250 mm × 4. 6 mm, 5μm), methanol (A)- 0. 1% phosphoric acid (B) was used as the mobile phase with gradient elution. The flow rate was 1.0 mL·min^-1, detective wavelength was 280 nm. The injection volume was 10 μL, column temperature was set at 30 ℃. Results The linearity of procyanidin B1 was 23.0-230 mg·L^-1 (r =0. 999 1) ,the average recovery was 96. 3% ;the linearity of procyanidin B2 was 14. 5- 145 mg·L^-1 (r =0. 999 4) ,the average recovery was 97.5 %. Conclusions This method is simple, accurate and reproducible for the determination of procyanidin B1 and B2 in grape seed extract.
