摘要 目的建立RP-HPLC法同时测定绵茵陈中绿原酸、3,5-二咖啡酰奎宁酸、4,5-二咖啡酰奎宁酸和金丝桃苷共4种有效成分的含量。方法采用RP-HPLC法。色谱柱为PhenomenexLunaC18柱(250mm×4.6mm,5μm),以乙腈-体积分数0.05%磷酸水为流动相,梯度洗脱,检测波长为345nm,流速1.0mL·min^-1,柱温30℃。结果绿原酸、3,5-二咖啡酰奎宁酸、4,5-二咖啡酰奎宁酸和金丝桃苷质量浓度分别在2.609-83.50mg·L^-1(r=0.9999)、1.853-59.31mg·L^-1(r=0.9996)、0.9480~30.34mg·L^-1(r=0.9998)和0.4506-14.42mg·L^-1(r=0.9999)内与峰面积呈良好的线性关系,平均加样回收率(n=9)分别为96.3%、101.2%、101.4%和96.5%。结论该方法准确可靠,可用于绵茵陈的质量控制。 Objective To establish an HPLC method for simultaneous determination of chlorogenic acid, hyperoside ,3,5-dicaffeoylquinic acid and 4,5-dicaffeoylquinic acid in Artemisiae Scopariae Herba (traditional Chinese medicine). Methods Isolation of sample was achieved on a Luna C18 column (250 mm×4.6 mm,5μm). A mixture of acetonitrile and 0. 05% phosphoric acid with gradient elution was used as mobile phase. The analytes were detected at 345 nm, and the column temperature was maintained at 30 ℃. Results The linearity ranges of chlorogenic acid, hyperoside, 3,5-dicaffeoylquinic acid, 5-dicaffeoylquinic acid were 2. 609- 83.50 mg· L ^- 1 ( r = 0. 999 9 ), 1. 853- 59.31 mg· L^ -1( r = 0. 999 6 ), 0. 948 0- 30. 34 mg· L^ -1( r = 0. 9998 ) and 0. 450 6- 14. 42 mg· L^ -1( r = 0. 999 9), respectively. The average recoveries ( n = 9) of them were 96. 3 %, 101.2%, 101.4% and 96. 5 %, respectively. Conclusions The method is simple, fast, sensitive and can be used for the quality control of Artemisiae Scopariae Herba.
