摘要
目的研究荨麻属植物宽叶荨麻不同提取物抑制5α-还原酶活性的有效部位。方法建立抑制5α-还原酶体外模型,从大鼠肝脏中提取5α-还原酶,与底物睾酮(testosterone,T)以及供氢体NADPH共同组成反应体系。采用高效液相色谱法,检测反应前后的睾酮含量,以睾酮的含量来判断5α-还原酶的活性。非那雄胺做阳性对照药。结果宽叶荨麻全草水提物的正丁醇部分具有显著的抑制5α-还原酶的活性。结论宽叶荨麻水提物的正丁醇萃取物为抑制5α-还原酶的活性的有效部位。
Objective To research the effective parts of Urtica plant extract for their inhibitory activity on 5α- reductase. Methods The in vitro 5α-reductase inhibitory model was established. The whole reactive system was composed of the 5α-reductase extracted from the livers of male rats,testosterone (as a substrate) and NADPH ( as a hydrogen supplier). The concentration of testosterone before and after the reaction was meas- ured by HPLC and the activity of 5α-reductase was tested by the content change of testosterone. Finasteride was used as a positive control. Results and Conclusions The result shows that the n-butanol fraction of the water extracts of Urtica laetevirens Maxim. could inhibit the activity of the 5α-redutase remarkably.
