摘要
本研究根据苹果锈果类病毒(ASSVd)基因组全序列设计LAMP特异性引物,并通过优化引物最终建立了ASSVd的RT-LAMP检测方法。该方法可有效区分同属的澳洲葡萄类病毒(AGVd)、葡萄黄点类病毒(GYSVd)、柑桔类病毒III(CiVd III),具有灵敏度高、特异性强及检测时间短的特点,可用于现场ASSVd的快速检测。
In this study, the pairs of specific primer were designed by analyzing Apple skar skin viroid (ASSVd) complete nucleotide sequence, and a loop-mediated isotermal amplification (RT-LAMP) method for the detection of ASSVd was established after the optimization of primers. This RT-LAMP method can effectively distinguished four Apscaviroid, Apple skar skin viroid (ASSVd), Australian grapevine viroid (AGVd), Grapevine yellow speckle viroid (GYSVd) and Citrus viroid III (CiVd III). The method has the characteristics of high sensitivity, strong specificity and rapidity, and can be used for the rapid detection of the filed samples.
