摘要
目的:通过检测动脉性勃起功能障碍(AED)大鼠转化生长因子β(TGF-β)及还原型辅酶Ⅱ(NADPH)的表达,观察中药疏肝益阳胶囊改善AED血管平滑肌功能的分子机制。方法:选取3月龄成年雄性SD大鼠,采用双侧髂内动脉结扎法制造勃起障碍模型。设假手术对照组、模型组、西药西地那非组(10.5mg·kg-1·d-1)、疏肝益阳小剂量治疗组(0.5g·kg-1·d-1)、大剂量治疗组(1g·kg-1·d-1),灌胃给药30d。荧光定量PCR法检测海绵体组织TGF-β、NADPH mRNA表达,ELISA法检测血浆TGF-β含量。结果:模型组与假手术组比较,TGF-β和NADPH表达显著升高(P<0.01);疏肝益阳治疗组与模型组比较,TGF-β和NADPH表达均显著降低(P<0.05)。结论:疏肝益阳胶囊对动脉性勃起障碍大鼠的血管平滑肌功能具有改善作用,而这种作用可能是通过调节氧化应激机制实现的。
Objective: To observe the molecular mechanism of the treatment of arteriogenic erectile dysfunction with Shugan Yiyang Capsule (SGYY) through the detection of TGF-βand NADPH. Methods: 60 three-month-old male Sprague- Dawley rats were included in the study. Bilateral ligation of the internal iliac artery was performed on rats in the experimental groups. All the rats were divided into control group, model group, sidenafil group, SGYY high-dosage group(1g.kg-1.d-1) and low-dosage group(0.5g.kg-1.d-1). After 30 days gavage, the plasma concentration of TGF-β in rats of each group was detected by ELISA, and expressions of TGF-β and NADPH mRNA in penis tissue of rats were detected by real-time PCR. Results: Expressions of TGF-β and NADPH were increased in model group compared to the control group (P〈0.01). And they were decreased in the SGYY treatment groups (P〈0.05). Conclusion: SGYY can improve the function of vascular smooth muscle in rat model with arteriogenic erectile dysfunction, and this effect may be as a result of regulating of oxidative stress mechanism with SGYY.
