柴疏四君汤对肝郁-脾虚-肝郁脾虚不同证候模型大鼠消化系统功能的影响 被引量:18

Influence of Chaishu Sijun Decoction on the changes in the digestive system of liver-qi stagnation-spleen-qi deficiency-liver-gistagnation and spleen-qi deficiency in rats

摘要 目的:比较肝郁、脾虚、肝郁脾虚三证模型大鼠消化系统的功能改变及柴疏四君汤对其影响。方法:大鼠随机分为正常对照、肝郁模型、肝郁给药、脾虚模型、脾虚给药、肝郁脾虚模型和肝郁脾虚给药共7组,分别采用慢性束缚、饮食失节+过度疲劳、慢性束缚+饮食失节+过度疲劳的方法制模。于造模第15天,给药各组大鼠按3.57g·kg-1·d-1给予柴疏四君汤灌胃;模型组和对照组予等量蒸馏水,连续14d。测定各组大鼠胃液中胃蛋白酶(Pepsin)、尿D-木糖排泄率(UD-xER)及血中胃动素(MTL)、胃泌素(GAS)、生长抑素(SS)、胆囊收缩素(CCK)。结果:与正常组大鼠比较,3个模型组UD-xER、CCK均显著性降低(P<0.05);其中脾虚组和肝郁脾虚组Pepsin显著性降低(P<0.05)、SS显著性升高(P<0.05),脾虚组和肝郁组MTL和GAS均显著性降低(P<0.05),肝郁脾虚组MTL和GAS显著性升高(P<0.05)。与各模型组大鼠相比,肝郁给药组GAS显著性升高(P<0.05);脾虚给药组UD-xER和CCK均显著性升高(P<0.01),SS显著性降低(P<0.05);肝郁脾虚给药组Pepsin、UD-xER及CCK均显著性升高(P<0.05),MTL、GAS及SS均显著性降低(P<0.05)。结论:肝郁、脾虚和肝郁脾虚三证模型大鼠均存在不同程度的胃肠功能异常,但同中有异;柴疏四君汤对三证模型大鼠的胃肠功能均有不同程度的改善作用,但以对肝郁脾虚证的作用最优。疏肝健脾方与肝郁脾虚证之间具有较高的关联。 Objective: To observe and compare the effects of Chaishu Sijun Decoction on the changes in the digestive system of liver-qi stagnation (GY)-spleen-qi deficiency (PX) -liver-qi stagnation and spleen-qi deficiency (GYPX) in rat. Methods: Rats were randomly divided into normal control group, GY group, GY medication group, PX group, PX medication group, GYPX group, GYPX medication group. Adopted by chronic restraint stress, excess fatigue + improper diet and chronic restraint stress + excess fatigue + improper diet to form the model. On the 15th day, medicated group rats were given Chaishu Sijun Decoction 1mL.g-1.d-1accordingly. Model group and normal group were given distilled water for 14 days. Detection of the activity of pepsin in gastric juice and the D-xylose excretion rate of urine and the content of motilin, gastrin, somatostatin and cholecystokinin in the blood of each model. Results: Compared with normal group, the three model groups declined markedly on the D- xylose excretion rate and the content of cholecystokinin (P〈0.05); The activity of pepsin of PX and GYPX had declined markedly (P〈0.05) and the content of somatostatin had inproved markedly (P〈0.05). The content of motilin and gastrin of GY and PX had declined markedly (P〈0.05), inproved markedly of GYPX models (P〈0.05). Compared with each model group, the medicated group rats were given Chaishu Sijun Decoction 3.57g/kg for 14days. the content of gastrin of the GY medication groups had inproved markedly (P〈0.05). The D-xylose excretion rate and cholecystokinin of PX medication groups had inproved markedly (P〈0.05), declined markedly in somatostatin. The activity of pepsin, D-xylose excretion rate and the content of cholecystokinin of GYPX medication groups had inproved markedly (P〈0.05), declined markedly in motilin, gastrin and somatostatin (P〈0.05). Conclusion: There are some similarities as well as some differences in the function of the digestive system of GY-PX-GYPX model in rat; Chaishu Sijun Decoction had effect on the function of the digestive system of GY-PX-GYPX model in rat, but it had better effect on the model ofGYPX syndrome. Chaishu Sijun Decoction had the markest relativity with GYPX syndrome.
