摘要
目的:研究硒对人乳腺癌裸鼠皮下移植瘤的生长及瘤体硒结合蛋白I(SBP1)表达的影响。方法:建立裸鼠皮下移植瘤模型,随机分成5组,即生理盐水组,5-氟脲嘧啶(5-FU)组,低、高剂量亚硒酸钠组,高剂量亚硒酸钠+5-FU组,动态监测各组裸鼠体质量及移植瘤的体积,免疫组织化学与免疫印迹检测移植瘤组织中SBP1蛋白的表达情况。结果:实验组裸鼠移植瘤体积与生理盐水组相比显著减小,实验组移植瘤组织中SBP1蛋白的表达比生理盐水组显著增加。结论:硒通过诱导SBP1的表达,对乳腺癌裸鼠移植瘤具有一定的治疗作用。
Objective: To study the expression and significance of selenium-binding protein 1 (SBP1) in breast carcinoma xenograft of nude mice treated with seleium, Methods: The model of xenotransplanted breast carcinoma was established in nude mice using the MCF-7 cell line. 40 mice were randomized into 5 groups: saline group, 15 mg/kg 5-FU group, 2 mg/kg Na2SeO3 group, 6 mg/kg Na2SeO3 group, 5-FU+6 mg/kg Na2SeO3 group. The weight of the nude mice and the volume of xenograft were measured in diferrent periods, SBP1 protein expression of xenograft was deetected by immunohistostaining and Western blotting. Results: The tumor volume of the nude mice in the experiment groups was significantly lower in comparison with those in the control group; immunohistostaining and Western blotting showed SBP1 protein expression of the experiment groups were significantly improved in comparison with the control group. Conclusion: The therapeutic effect of seleium on breast carcinoma xenograft in nude mice may be mediated by SBP1 induction.
