摘要
目的探讨原花青素(PC)对As2O3所致雄性小鼠的生殖系统氧化损伤的拮抗作用。方法选用昆明种雄性小鼠60只,随机分配到对照组,As2O3组,PC组,砷+PC低、中、高剂量组;As2O3按4.0 mg/kg·bw染毒剂量灌胃;PC按100、200和400 mg/kg剂量灌胃;连续染毒5周后,取其睾丸,匀浆后检测氧化损伤指标丙二醛(MDA)、8-OHdG,抗氧化指标T-AOC、谷胱甘肽(GSH)、超氧化物歧化酶(SOD)。采用2×2析因设计和单因素方差分析方法进行分析。结果As2O3组小鼠MDA(5.13±0.62)nmol/mg prot、8-OHdG(0.75±0.05)ng/ml含量均高于对照组(2.71±0.48 nmol/mg prot,0.61±0.04 ng/ml);PC对As2O3染毒小鼠的MDA(F=10.662,P=0.003)、8-OHdG(F=31.651,P<0.001)、GSH(F=23.114,P<0.001)的含量交互作用具有统计学意义。中剂量PC组MDA(3.14±0.47)nmol/mg prot、8-OHdG(0.72±0.05)ng/ml显著高于As2O3组,GSH(32.27±6.13)mg/mg prot显著高于As2O3组;高剂量PC组MDA(3.43±0.39)nmol/mg prot(F=28.506,P<0.001)、8-OHdG(0.68±0.03)ng/ml(F=5.439,P=0.003)显著低于As2O3组,GSH(38.37±5.91)mg/mg prot(F=5.086,P=0.005)显著高于As2O3组。结论 PC对As2O3致雄性小鼠生殖系统的氧化损伤具有拮抗作用,中高剂量PC抗氧化损伤效果较好。
Objective To observe the antagonism of proanthocyanidin (PC) on arsenic trioxide (As2O3 )-induced the oxidative damage of the genital system in male miee. Methods 60 KM male mice were randomly divided into 6 groups, including control, As2O3 , PC, As2O3 + lower dose PC, As2O3 + medium dose PC, As2O3 + high dose PC. The mice were administrated intragastrieally with the dose of As2O3 4. 0 mg/kg.bw and with the dose of PC 100 mg/kg,200 mg/kg,400 mg/kg. After 5-week treatment on the male mice, the testis were taken out to prepare for the homogenate. We detect the concentrations of MDA, 8-OHdG, T-AOC, GSH, SOD as the index of the oxidative damage and ant-oxidation. The 2 ×2 factorial design ANOVA were used to analyze the effect. Results The concentration of MDA(5. 13 ± 0. 62) nmol/mgprot,8-OHdG(0.75 ± 0.05 ) ng/ml of As203 group were higher than that of the control group ( 2.71 ± 0.48 nmol/mgprot, 0.61 ±0.04 ng/ml). The interaction between PC and As203 on MDA(F = 10. 662,P =0. 003) ,8-OHdG(F =31. 651,P 〈0. 001) ,GSH(F = 23. 114,P 〈 0. 001 ) were significant. The concentration of MDA ( 3.14 ± 0.47 ) nmol/mg prot, 8-OHdG ( 0.72± 0.05 ) ng/ml of the medium dose of PC were significantly lower than that of As203 group. While, the concentration of GSH (32.27 ± 6.13 ) mg/mg prot was higher than that of As2 03 group. The concentration of M DA ( 3.43±0. 39 ) nmol/mg prot ( F = 28. 506, P 〈 0. 001 ), 8-OHdG (0. 68 ±0. 03 ) ng/ml(F = 5. 439,P =0. 003) of the high dose of PC were significantly lower than that of As203 group. And, the concentration of GSH (38.37 ± 5.91 ) mg/mg prot ( F = 5. 086, P = 0. 005 ) was higher than that of As2 03 group. Conclusion PC has protection effects on the oxidative damage of genital system in male mice induced by As203. The medium and high dose of PC has more significant anti-oxidation.
