ARHI基因mRNA和蛋白表达水平与胶质瘤恶性度相关性探讨 被引量:2

摘要 目的探讨ARHI基因表达水平与人脑胶质瘤恶性度的相关性。方法采用RT-PCR和Western blot法检测10例正常脑组织、59例胶质瘤组织、4种胶质瘤细胞系(U251,SHG44,BT325,U87)中ARHI基因mRNA和蛋白的表达强度,结合临床病理资料统计分析。结果胶质瘤组织、细胞系中ARHI基因的表达水平显著低于正常脑组织,胶质瘤组织中ARHI的表达水平随肿瘤恶性程度增加而明显降低(P<0.01)。结论 ARHI基因mRNA和蛋白在胶质瘤中低表达,且表达水平与胶质瘤恶性程度负相关,提示ARHI基因低表达对胶质瘤的形成和恶性演变具有重要作用。 Objective To investigate the relationship between ARHI expression and malignant grades of human brain glioma . Methods RT-PCR and Western blot methods were used to detect the ex-pression level of mRNA and protein of ARHI gene in 59 pairs of giloma samples ,10 normal brain tissues and 4 malignant glioma cell lines(U251 ,SHG44 ,BT325 and U87) . The implications between the expres-sion of ARHI and clinical-pathological factors were analyzed . Results It was found that the expression of ARHI in glioma tissues and glioma cell lines were significantly lower than that in normal brain tissues . The expressions of ARHI mRNA and protein decreased as the upgrading of glioma malignant grade fromⅠ to Ⅳ(P〈0 .01) . Conclusions The low expressions of ARHI mRNA and protein were founded in gli-oma and the expression levels were negatively correlated with the malignant grade of glioma . It was indi-cated that the low expression of ARHI might contribute to the occurrence and malignant transformation of glioma .
